O78血清型禽致病性大肠杆菌油乳剂灭活疫苗的研制及其对鸭的免疫保护作用

【摘要】 为有效控制禽大肠杆菌病的流行,制备了O78优势血清型禽致病性大肠杆菌(APEC)油乳剂灭活疫苗,并评价它对鸭的免疫保护效果。通过体内复壮、半数致死量测定、生物学特性分析筛选制备疫苗用菌株。然后制备油乳剂灭活疫苗,并测定其免疫保护率、最佳免疫剂量筛选、最佳免疫次数、免疫持续期及交叉保护性。结果显示,APCE94菌株制备的疫苗免疫保护性好、安全性高,可作为疫苗候选株。该疫苗仅以每只5×108 CFU的免疫剂量免疫1次即可提供90%的保护率,免疫雏鸭60d后仍可以提供很好的保护作用。本研究制备的O78血清型APEC油乳剂灭活疫苗可以有效保护鸭不感染O78血清型APEC,有助于鸭大肠杆菌病的防控治疗。

【Abstract】 To control effectively avian colibacillosis,inactivated oil-emulsion vaccine against O78 serotype avian pathogenic Escherichia coli(APEC)was developed and evaluated in ducks.We selected the vaccine candidate strains by rejuvenation in vivo,virulence determination and biological characteristics analysis.Then immunization dose and times,protection rate and duration of the vaccine were investigated.The results showed that APCE94 strain provided the best safety and immune protection to duck.Each duck received the vaccine of one time at a dose of 5×108 CFU can provide 90% protection level,and the good protection was up to 60 days post-immunization.However,this inactivated vaccine against O78 serotype APEC could not provide cross-protection to duck for O1 and O2serotype after APEC infection.In conclusion,we developed an inactivated oil-emulsion vaccine against O78 serotype APEC,which can effectively protect ducks from the O78 serotype APEC infection and will be help to prevent and control duck colibacillosis.
