不同形态硒、维生素E、紫胡萝卜提取物单独和联合使用对H2O2诱导H9c2心肌细胞损伤的保护作用
The Effects of Individual and Combined Selenium, Vitamin E and Anthocyanin Extracts of Purple Carrots against the H2O2-induced Oxidative Stress in H9c2 Cells
【摘要】 目的:观察不同形态硒(L-硒甲基硒代半胱氨酸、亚硒酸钠、富硒酵母)、维生素E、紫胡萝卜提取物(主要成分为花青素)单独和联合用药对H2O2诱导H9c2细胞损伤的保护作用,并探讨其作用机制。方法:将H9c2细胞随机分为11组,即1空白对照组,2H2O2模型组,3L-硒甲基硒代半胱氨酸组(Se MSC),4亚硒酸钠组(SS),5富硒酵母组(SEY),6维生素E组(VE),7紫胡萝卜提取物组(Ant),8VE+Ant联用组,9Se MSC+VE+Ant联用组,10SS+VE+Ant联用组,11SEY+VE+Ant联用组。经不同样品组干预处理后,通过H2O2诱导细胞氧化损伤,检测各组H9c2细胞的存活率,脂质氧化终产物丙二醛(MDA)含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活力。结果:与模型组相比,不同形态硒、维生素E、紫胡萝卜提取物单独和联合用药组H9c2细胞的存活率明显增加。同时,细胞内MDA的含量降低,SOD、CAT、GSH-Px的抗氧化活力增强,联合用药组的效果优于单独用药组。结论:不同形态硒(L-硒甲基硒代半胱氨酸、亚硒酸钠、富硒酵母)、维生素E、紫胡萝卜提取物均可保护由H2O2诱导引起的H9c2细胞氧化损伤,提高心肌细胞抗氧化能力,联合用药组的效果优于单独用药组,具有明显的抗氧化协同作用。
【Abstract】 Objective: in order to research the effects of individual and combined selenium, vitamin E(VE), anthocyanin extracts of purple carrots against the H2O2-induced oxidative stress in H9c2 cells, and the possible mechanism.Methods: H9c2 cells were randomly divided into 11 groups: 1Control group(control), 2H2O2 model group(model),3L-Se-Methylselenoeysteine group(Se MSC), 4Sodium selenite group(SS),5Selenium-enriched yeast group(SEY), 6Vitamin E group(VE), 7Anthocyanin extracts of purple carrots group(Ant), 8VE+Ant combination group, 9Se MSC+VE+Ant combination group, 10SS+VE+Ant combination group 11SEY+VE +Ant combination group. After incubated with sample of each group for 24 h, H9c2 cells were treated by 400 umol/L H2O2 for 6 h. Cell viability was evaluated by MTT assay and malonaldehyde(MDA) content, superoxide dismutase(SOD), catalase(CAT) and glutathione peroxidase(GSH-Px) activities were measured. Results: compared with model group, the individual and combined treatments of selenium, vitamin E, anthocyanin extracts could increase the viability of H9c2 cell significantly. Moreover, the MDA contents in cells were decreased and the activities of SOD, CAT, GSH-Px were increased by pretreated with the samples.The changes of the combined groups were better than those of individual groups. Conclusion: different forms of selenium(L-Se-Methylselenoeysteine, sodium selenite, selenium-enriched yeast), vitamin E, anthocyanin extracts of purple potato could significantly improve the antioxidant activity against H2O2-induced injury in H9c2 cells. In additional, the combined groups showed greater antioxidant activity than the individual groups in protecting the oxidative damage of H9c2 cells.
