脂肪细胞因子抵抗素对卵巢癌细胞增殖影响及其机制研究
Mechanism of proliferation promotion by resistin in ovarian epithelial cancer cell
【摘要】 目的探讨脂肪细胞因子抵抗素(resistin)对卵巢癌细胞系HO-8910细胞增殖的影响及初步表达机制。方法 2011年5-10月于中国医科大学中心实验室分别采用噻唑蓝(MTT)比色法,检测不同浓度的抵抗素对卵巢癌HO-8910细胞增殖能力的影响;利用si RNA敲除脂肪细胞中的抵抗素,转移条件培养基培养卵巢癌细胞HO-8910,通过MTT法检测细胞增殖情况;利用免疫印迹法(western blot)检测抵抗素作用于HO-8910细胞后m TOR及其下游靶蛋白S6总蛋白的磷酸化蛋白量的变化。结果抵抗素可以呈时间、剂量依赖性地促进HO-8910细胞的增殖,30~100μg/L抵抗素作用24h即可以显著促进HO-8910细胞的增殖,48h的作用更为明显(P<0.05);脂肪细胞条件培养基可显著促进HO-8910细胞的增殖,如果在脂肪细胞培养过程中给予特异性敲低抵抗素,则作用反之;抵抗素作用卵巢癌HO-8910细胞24h后m TOR及其下游靶蛋白S6的磷酸化水平显著增加。结论抵抗素对人卵巢癌细胞系HO-8910细胞的增殖能力有明显的促进作用,该作用机制可能与m TOR信号通路有关,可为诊治卵巢癌提供新的靶点。
【Abstract】 Objective To investigate the resistin effect on proliferation of ovarian epithelial cancer cell HO-8910 and its mechanisms.Methods The proliferation of HO-8910 was examined by MTT assay;HO-8910 cells were transfected with the resistin interfering RNA(si RNA);si RNA was wsed to knock out the resistin in adipocytes;transfer conditions medium was used to culture ovarian cancer HO- 8910 cells;apply MTT was used to detect the proliferation of the cell;m TOR protein of S6 and phosphorylation of S6 was measured by western blot after providing resistin.Results Resistin at concentration of 30~100 μg/L promoted the proliferation of ovarian epithelial cancer cell HO- 8910 in a time and dose-dependent manner;the effect of resistin at 100μg/L for 48 h was most prominent(P<0.05);resistin promoted the proliferation and resistin si RNA significantly attenuated the proliferation in the HO-8910-adipocytes-transfer medium.Conclusion The resistin may play important roles in ovarian cancer cell proliferation and m TOR can promote the proliferation of HO-8910 cells,and may be regarded as a new targetor in the diagnosis and treatment of ovarian cancer.
