活细胞在线检测器在长双歧杆菌BBMN68培养中的应用研究

【摘要】 活菌数检测是益生菌培养过程分析的重要环节。传统以平板稀释法检测益生菌活菌数费时费力,且无法实现实时监测。本文探索了电容型活细胞在线检测器在双歧杆菌BBMN68培养过程中的应用效果,评价其检测活细胞量的可行性。研究表明,在菌体发酵生长的对数期,活细胞在线检测器的电容值与长双歧杆菌BBMN68活细胞量之间存在良好的线性关系,其线性相关系数为0.992,比吸光值法更准确地反映发酵体系中活细胞量的情况,在稳定期与衰亡期也表现出了良好的相关性。比平板计数法更快速,可以实现实时监控。电导型活细胞检测器为益生菌培养过程在线检测提供了一种简便、精确的方法。

【Abstract】 An on-line viable-cell mass sensor was used to monitor the Viable cells of Bifidobacterium longum BBMN68 fermentation process. In this paper, this new method that based on the capacitive principle was employed. As a result, the relationship between the number of colony forming units(CFU) and capacitance with a correlation coefficient(R) of 0.992 was achieved and data showed a good reproducibility under the same culture conditions. Compared with the results determined by absorbency method, the capacitive method is more reliable. Therefore, it could be concluded that a capacitance probe was a simple as well as a accurate tool in Bifidobacterium longum BBMN68 cultivation process.
