一株鹅圆环病毒的全基因组测序及序列分析

【摘要】 从山东省潍坊地区某朗德鹅鹅场的发病鹅体内检测到1株鹅圆环病毒,命名为SDWF-01。根据Gen Bank上已发表的鹅圆环病毒序列,设计了一对特异性引物,利用PCR技术对该株病毒进行全基因组克隆并测序,对其基因组核苷酸组成、结构及遗传进化特性进行分析。结果表明:SDWF-01株全长1 821 bp,编码4个开放阅读框;与Gen Bank上已发表的鹅圆环病毒核苷酸序列同源性为91.4%~98.5%,其中SDWF-01株与yk3毒株的同源性最高(98.5%),而与浙江株xs5的同源性最低(91.4%)。研究首次对山东地区鹅圆环病毒进行检测,并完成全基因组克隆测序及核苷酸序列分析。

【Abstract】 A goose circovirus(Go CV)strain,named SDWF-01,was detected from Landes geese farm in Shandong province. A pair of specific primers was designed based on sequences of circovirus published on Gen Bank,the complete genomic sequence was amplified using PCR,then molecular structure and genetic characteristics were investigated. The results showed that the whole genome sequence of SDWF-01 strain had a total length of 1 821 nucleotides,consisting of 4 open reading frames;The nucleotide homology of genomic sequences between SDWF-01 strain and other Go CV isolates were ranged from 91.4% to 98.5%,SDWF- 01 strain shared a high identity of 98.5% with yk3 strain(DQ192280.1)and a low identity of 91.4% with xs5 strain(DQ192287.1). It was the first report on the detection and full genome sequencing of Go CV in Shandong Province.
