Skp2和p27Kip1表达异常与乳腺癌发生及临床病理特征的相关性研究

【摘要】 目的从蛋白水平研究Skp2和p27Kip1表达异常与乳腺癌发生及临床病理特征的相关性。方法采用En Vision两步法检测30例乳腺正常组织,30例导管上皮不典型增生组织(ADH)、30例导管原位癌(DCIS)和56例浸润性导管癌(非特殊类型)(IDC)中Skp2和p27Kip1的表达水平,并分析Skp2和p27Kip1表达异常与乳腺癌发生及临床病理特征的关系。结果 1ADH组、DCIS组及IDC组的Skp2阳性率均高于正常对照组(P<0.05,P<0.01,P<0.01),IDC组和DCIS组高于ADH组(P<0.01,P<0.05),IDC组又高于DCIS组(P<0.01)。ADH组、DCIS组和IDC组p27kip1阳性率均低于正常对照组(P<0.05,P<0.01,P<0.01),IDC组和DCIS组均低于ADH组(P<0.01,P<0.05),IDC组又低于DCIS组(P<0.01)。2IDC伴淋巴结转移组Skp2阳性率显著高于DCIS淋巴结转移组(P<0.05);IDCⅢ级高于IDCⅠ~Ⅱ级(P<0.01)。反之,p27kip1阳性率在IDC伴淋巴结转移组低于无淋巴结转移组(P<0.05);IDCⅢ级低于Ⅰ~Ⅱ级(P<0.01)。结论 Skp2与p27Kip1异常表达与乳腺癌发生、病理分级和淋巴结转移有密切相关性。

【Abstract】 Objective To explore the relationship of Skp2 and p27Kip1 with pathological feature and carcinogenesis of breast. Methods The positive expression rates of Skp2 protein and p27kip1 protein were detected in 30 cases of normal control group,30 cases of atypical ductal hyperplasia( ADH),30 cases of ductal carcinoma in situ( DCIS) and 56 cases of invasive ductal carcinoma( IDC). Results First,the expression of Skp2 and p27kip1 in different pathological groups ADH,DCIS and IDC was higher than that in normal control group( P < 0. 05,P < 0. 01,P < 0. 01); in IDC group,the positive expression rate of Skp2 protein in DCIS group was higher than that of ADH group( P < 0. 01,P < 0. 05); that in IDC was higher than that of DCIS( P < 0. 01). P27kip1 protein levels in ADH,DCIS and IDC were lower than that of normal control group( P < 0. 05,P < 0. 01,P < 0. 01); that in IDC and DCIS were lower than that in ADH group( P < 0. 01,P <0. 05). P27kip1 expression in IDC was lower than that of DCIS( P < 0. 01). Second,the protein levels of Skp2 and p27kip1 varied in different pathological grade and lymph node metastasis group. In IDC with lymph node( +) group,Skp2 positive expression rate was significantly higher than that of lymph node(-) group( P < 0. 05); that in IDC with grade III was higher than that of grade I-II( P < 0. 01). On the contrary,p27kip1 positive expression rate in IDC with lymph node metastasis( +) group was lower than that of lymph node metastasis(-) group( P < 0. 05); P27kip1 positive expression rate of IDC with grade III was lower than that of grade I-II( P < 0. 01). Conclusion The abnormal expression of Skp2 and p27Kip1 is closely correlated with pathologic stage and lymph node metastasis in breast cancer.
