【摘要】 目的应用非标记定量技术研究特应性皮炎患儿尿液中的差异表达蛋白,筛选特应性皮炎患儿尿液中的特异性蛋白质标记物。方法收集特应性皮炎患儿和健康对照者的尿液各6份,处理和提取尿液蛋白质,经一维SDS-PAGE和蛋白胶内酶解获得肽段,应用高效液相色谱串联质谱鉴定,使用Mascot软件查询并用Scaffold软件作相对定量分析。结果特应性皮炎患儿组和健康对照组的尿液共检测出103种表达差异倍数大于1.5倍的蛋白质,患儿组表达升高的有48种蛋白质,表达降低的有55种蛋白质,其中10种蛋白质仅在患儿组检测到,17种蛋白质仅在对照组检测到。结论特应性皮炎患儿与正常儿童尿液间存在蛋白质表达差异。更多还原

【Abstract】 Objective Label-free quantitative proteomics method was applied to find differentially expressed urinary proteins in patients with atopic dermatitis. Urinary protein markers for atopic dermatitis were screened.Methods Six urinary samples from atopic dermatitis patients and normal controls were treated,and the proteins were extracted. The proteins were separated with One-dimensional SDS-PAGE electrophoresis,and the peptides were obtained from the gel after enzymolysis. High performance liquid chromatography tandem mass spectrometry identification was performed. Mascot and scaffold software were used to search and relatively quantitate the protein. Results The content of 103 proteins in the urine varied more 1. 5folds between atopic dermatitis patients and normal controls,including 48 up-regulated and 55 down-regulated proteins in atopic dermatitis patients. Among them,10 proteins were only detected in patients’ urine.17 proteins were only in normal controls. Conclusion The expression of urinary proteins is different between atopic dermatitis and normal children.更多还原
