前列通窍胶囊对前列腺增生大鼠模型的影响

【摘要】 目的探讨前列通窍胶囊对前列腺增生(BPH)大鼠模型的影响,初步探讨其作用机制。方法将60只大鼠随机取10只作为空白组,其余50只大鼠采用去势、皮下注射丙酸睾酮法造成前列腺增生模型。将常规造模成功的50只大鼠随机分为模型组、癃闭舒胶囊对照组和前列通窍胶囊低、中、高剂量组,每组10只。空白组、模型组给予蒸馏水,其他各组分别予以相应药品混悬液灌胃给药,给药30天后处死大鼠,称取前列腺湿重并计算前列腺指数,测定前列腺组织中睾酮(T)、雌二醇(E2)的含量及表皮生长因子(EGF)的水平,测定前列腺的腺体密度并用光镜观察前列腺组织病理学改变。结果与模型组相比,前列通窍胶囊各剂量组均可一定程度降低前列腺增生模型大鼠前列腺湿重及前列腺指数,高、中剂量组可显著降低大鼠前列腺的腺体密度(P<0.01),均可显著降低T、EGF水平(P<0.01),但对E2的影响不明显。结论前列通窍胶囊对前列腺湿重及前列腺的腺体密度均有一定影响,能显著降低前列腺组织中T、EGF水平,揭示了前列通窍胶囊治疗BPH的部分作用机制。

【Abstract】 Objective: To study the effect of Qianlie Tongqiao Capsule on benign prostate hyperplasia rat models( BPH) and to explore its mechanism. Methods: 60 rats were randomly assigned to take 10 as a control group,and the remaining 50 rats were castrated,causing BPH models by subcutaneous injection of testosterone propionate. The conventional 50 successful modeled rats were randomly divided into a model group,aRongbishu capsule control group and low,medium and high dose groups of Qianlie Tongqiao Capsule,10 mice per group.The non- drug group and the model group were given distilled water while the other groups were given intragastric administration with corresponding drugs. The rats were sacrificed after 30 days to take their prostate to weigh their wet weight and calculate their index,to measure their testosterone( T),estradiol( E2) and epidermal growth factor( EGF) level in prostate tissue as well as the density of prostate gland. The light microscope was used to observe their pathological changes. Results: Compared with the model group,each group with different doses of Qianlie Tongqiao Capsule could reduce the prostate wet weight and prostate index of prostatic hyperplasia rat to some extent.The high dose group could significantly reduce the density of the rat prostate gland( P < 0. 01) and T,EGF levels( P < 0. 01),but no obvious effect on the E2. Conclusion: Qianlie Tongqiao Capsule has a certain influence on the prostate wet weight and prostate gland density and can significantly reduce prostate tissue T,EGF level. The study reveals some mechanism of Qianlie Tongqiao Capsule for BPH treatment.
