正交实验优化鱼类保肝中药复方的提取工艺

【摘要】 研究影响鱼类保肝中药复方提取效果的各种因素,确定最佳提取工艺。运用单因素试验考察溶剂倍量(A)、浸泡时间(B)、煎煮时间(C)和煎煮次数(D)4个因素对中药复方中君药丹酚酸B含量的影响。采用正交实验法,对水煎煮工艺条件进行优选。在所考察的4个因素中,对中药复方提取效果的影响程度依次为:C>A>D>B,即煎煮时间>溶剂倍量>煎煮次数>浸泡时间,确定最佳水煎煮工艺为A2B1C1D2,即10倍水量,浸泡0.5 h,煎煮时间为0.5 h,煎煮2次。本研究优选工艺设计合理,稳定可行,可为鱼类保肝药物的开发提供参考。

【Abstract】 To determine the best extraction technique,the effects of different factors on the extraction of traditional Chinese medicine compound for fish liver were studied.Single factor experiments were employed to observe the influences of the amount of added water(A),the time of soaking(B),decoction time(C),and decoction times(D) on the yield of salvianolic acid B.The orthogonal test method was used to optimize the extracting process of the water decoction.The effects of the four factors were listed as follows:C > A > D > B.The best extraction technique was described as A2B1C1D2.The best extracting process of water decoction was as follows;Adding 10 times water,soaking for 0.5 h,decocting 2 times and decocting for 1 h per time.The optimized process can be used for extracting salvianolic acid B from traditional Chinese medicine compound due to its stable,reasonable and feasible.
