miR-21在早、晚期肺癌及肺大疱患者血清中的表达及临床意义

【摘要】 目的探讨miR-21在早、晚期肺癌及肺大疱患者血清中的不同表达及其临床意义。方法以早、晚期肺癌患者及肺大疱患者(每组各20例,共60例)治疗前血清作为研究对象,使用RT-PCR检测其血清中miR-21表达变化,ROC曲线评估其敏感度和特异性。结果早、晚期肺癌患者血清中miR-21表达均明显高于肺大疱患者(P=0.003和P=0.000),且晚期肺癌患者血清中miR-21表达要高于早期肺癌患者(P=0.012)。miR-21诊断早、晚期肺癌的敏感度和特异性分别为65%和85%(AUC=0.775)、95%和85%(AUC=0.928);miR-21用于区别肺癌晚期与早期的敏感度、特异性分别为45%和95%(AUC=0.733)。结论人血清miR-21表达检测可能有助于肺癌的早期诊断及判断其分期。

【Abstract】 Objective To identify circulating miR-21 expression in human serum as a biomarker for early detection and stratification in lung cancer.Methods We analyzed serum levels of miR-21 in 20 lung cancer patients with stage Ⅰ,20 patients with stage Ⅲor Ⅳ,and 20 patients with pulmonary bulla.Blood samples were collected before surgery and therapy.Quantitative reverse transcription-polymerase chain reaction of circulating miR-21 was performed.Receiver operating characteristic analysis was used to evaluate the sensitivity and speci? city of serum miR-21.Results We found that serum miR-21 was consistently expressed in both early and advanced stage lung cancer patients while in advanced stage patients which were more significantly.miR-21 expressions were capable of separating early and advanced stage lung cancer patients from control groups with an area under the curve of 0.775 and 0.928,separately.Conclusion Serum miR-21 may represent a novel biomarker for early detection of lung cancer and a potentially useful biomarker for disease progression.
