γ-生育三烯酚与5-氟尿嘧啶联用对人胃癌细胞SGC-7901的增殖抑制作用
Antiproliferative Effect of Combined Treatment with γ- Tocotrienol and 5-FU on Gastric Cancer SGC-7901 Cells
【摘要】 目的离体条件下研究γ-生育三烯酚(γ-tocotrienol,γ-T3)与5-氟尿嘧啶(5-FU)联合应用对人胃癌细胞SGC-7901增殖抑制的作用方式。方法采用CCK-8法用不同浓度5-FU(0、30、50、70、90、110μmol/L)、不同浓度γ-T3(0、5、10、20、30、40μmol/L)以及不同浓度5-FU(0、30、50、70、90、110μmol/L)联合γ-T3(20或30μmol/L)分别作用于对数生长期的SGC-7901细胞48h,测定各药物对SGC-7901细胞增殖抑制率,并通过两药相互作用系数(coefficient of drug interaction,CDI)值[1]判定5-FU和γ-T3联合作用效果。用AnnexinⅤ-FITC/PI双染的流式细胞术法分别检测5-FU(30μmol/L)和γ-T3(20μmol/L)单独应用及两药联合应用48h诱导SGC-7901细胞凋亡情况。结果 5-FU和γ-T3均可抑制SGC-7901细胞的增殖,并随着浓度的增加抑制作用增强(P<0.05),增殖抑制作用呈明显的剂量效应关系;而联合应用与对照组和单药组相比对SGC-7901细胞的增殖抑制作用增强,有增效作用,其差异有统计学意义(P<0.05);流式细胞术凋亡结果表明,5-FU和γ-T3均可诱导SGC-7901细胞发生凋亡的作用(P<0.05),5-FU和γ-T3联合较单独应用,诱导细胞凋亡的作用增强(P<0.05)。结论γ-T3与5-FU联合用药与单独用药相比对人胃癌细胞SGC-7901的增殖抑制及诱导凋亡有明显的增效作用。
【Abstract】 Objective To investigate the suppression of cell proliferation of SGC- 7901 gastric cancer cells by combination γ- tocotrienol( γ- T3) with 5- FU in vitro. Methods The effect of γ- tocotrienol( 0,5,10,20,30,40μmol / L) and 5- FU( 0,30,50,70,90,110μmol / L) on proliferation of gastric cancer cell lines for 48 h was examined by Cell Counting Kit- 8( CCK- 8). To determine if the combined effect of γ- tocotrienol( 20 or 30μmol / L) and 5- FU( 30μmol / L) is synergistic,we calculated the value of coefficient of drug interaction( CDI). The apoptosis assay were tested by flow cytometric analysis. Results The treatment with γ- tocotrienol or 5-FU alone induced a significant dose- responsive growth inhibition( P < 0. 05),whereas combination treatment with these agents synergistically inhibited the growth of SGC- 7901 cells( P < 0. 05). The results of flow cytometric analysis showed that the treatment with γ- tocotrienol or 5- FU alone initate apoptosis( P < 0. 05),and combination treatment had more significant effect in apoptosis than these agents alone( P < 0. 05). Conclusion The finding indicates that γ- tocotrienol can potentiate the effects of antiproliferative and apoptosis- inducing in SGC- 7901 cells treatment by 5- FU.
