检测植物倍性时流式细胞仪参数设置的体会
Experience of flow cytometry parameter setting about ploidy analysis in plant
【摘要】 流式细胞仪已广泛用于植物倍性的检测,通过检测拟南芥叶片的倍性,对BD LSRFortessaTM型流式细胞仪在用于植物倍性检测时的参数选择和设置进行了研究。结果表明:当建立双参数FSC/SSC散点图圈定目标植物细胞核时,设定FSC-A、SSC-A为对数放大;以DAPI为"阈"进行阈值的设定,能有效地排除细胞碎片等大量非目标细胞核和噪音的干扰信号,且最大限度地保留了靶细胞核;建立DAPI-W/DAPI-A双参数散点图能有效排除黏连细胞核。
【Abstract】 Flow cytometry( FCM) for ploidy analysis of plant is a rapid and accurate measurement.BD LSRFortessaTMFCS was used to analyze the ploidy of Arabidopsis thaliana. To circle the target plant cell nucleus through establishing the FSC / SSC scatter diagram,it is required to set logarithmic amplifier of FSC-A and SSC-A. In order to remove the interference signal of the cell debris and noises,DAPI is selected as the threshold. To exclude the adhesion nucleus,the DAPI-W / DAPI-A scatter diagram is necessary to be establishing.
