LC-MS/MS法测定血浆氯吡格雷活性代谢产物衍生物的浓度
Determination of plasma concentration of active metabolite derivatives of clopidogrel by LC-MS/MS
【摘要】 目的建立液相色谱/质谱(LC-MS/MS)技术测定血浆中氯吡格雷活性代谢产物衍生物(CAMD)浓度的方法。方法色谱柱为Sapphire C18(250mm×4.6mm,5μm),柱温40℃;流动相为乙腈-0.1%甲酸(95∶5),流速为1ml/min;采用电喷雾正离子化三重四级质谱检测。结果 CAMD的回归方程为Y=0.01734X+0.00001318(r=0.997),线性范围为0.50-100ng/ml,相对回收率为98.4%-105.0%,绝对回收率为96.7%-120.2%,准确度、精密度及稳定性良好。结论 LC-MS/MS技术能快速、准确地测定血浆中CAMD浓度。
【Abstract】 Objective To establish a liquid chromatography-tandem mass spectrometry(LC-MS/MS)method for the determination of plasma concentration of active metabolite derivatives of clopidogrel(CAMD).Methods Sapphire C18column(250mm×4.6mm,5μm)was used at 40℃.The mobile phase consisted of a mixture of acetonitrile-0.1% formic acid(95∶5)in a flow rate of 1ml/min.The mass spectrometer was conducted in positive electrospray ionization mode.Results The regression equation for CAMD was Y=0.01734X+0.00001318(r=0.997)with the linear range of 0.50-100ng/ml.The relative and absolute recovery rates were 98.4%-105.0% and 96.7%-120.2%,respectively.Besides,the accuracy,precision and stability of CAMD were good.Conclusion LC-MS/MS method is accurate,sensitive and suitable for detecting plasma concentration of CAMD.
