乌司他丁对急性肺损伤大鼠肺TLR4及TREM-1和TREM-2表达的影响

【摘要】 目的探讨乌司他丁对急性肺损伤(ALI)大鼠肺组织中Toll样受体4(TLR4)及肺泡灌洗液中的髓样细胞触发受体1(TREM-1)和TREM-2表达的影响。方法将72只SD大鼠随机均分为三组。LPS组、LU组大鼠腹腔注射脂多糖(LPS)5mg/kg建立ALI模型;C组腹腔注射等容量生理盐水。LU组同时腹腔注射乌司他丁10万U/kg。每组给药后4、8、12和24h分别处死6只大鼠,采用RT-PCR半定量检测肺组织TLR4mRNA表达,ELISA法检测肺泡灌洗液中TREM-1和TREM-2表达。结果 LPS组各时间点的TLR4 mRNA、TREM-1和TREM-2表达均高于C组(P<0.05);LU组TLR4mRNA和TREM-1表达均低于LPS组(P<0.05),而TREM-2表达高于LPS组(P<0.05)。结论乌司他丁对LPS诱导的ALI有保护作用,可能与下调TLR4 mRNA、TREM-1表达和上调TREM-2表达有关。

【Abstract】 Objective To investigate the effects of ulinastatin on the expressions of Toll-like receptor 4(TLR4),triggering receptor expressed on myeloid cells 1(TREM-1)and TREM-2in lung tissues of rats with acute lung injury(ALI).Methods Seventy-two SD rats were equally and randomly divided into 3 groups of LPS,LU and C.The rats in groups of LPS and LU were intraperitoneally injected with lipopolysaccharide(LPS)5mg/kg to establish ALI models,which of group LU were given additional intraperitoneal injection of ulinastatin 100 000U/kg and of group C were given same volume of normal saline as the controls.At 4,8,12 and 24hours after administration,6rats in every group were sacrificed and the expressions of TLR4 mRNA in lung tissues and TREM-1and TREM-2in bronchoalveolar lavage fluids were detected by semiquantitative RT-PCR and ELISA,respectively.Results The expressions of TLR4 mRNA,TREM-1and TREM-2 were all higher in group LPS than those in group C(P<0.05).The expressions of TLR4 mRNA and TREM-1 were lower,but TREM-2 expression was higher,in group LU than those in group LPS(P<0.05).Conclusion Ulinastatin has a protective effect on ALI induced by LPS in rats,which may be related to downregulating the expressions of TLR4 mRNA and TREM-1and upregulating the expression of TREM-2.
