【摘要】 目的研究基质金属蛋白酶9(MMP-9)在C端的血红素样结构域(MMP-9-PEX)和MMP-9在垂体腺瘤侵袭性生长中的作用及其相关性。方法用RT-PCR法检测116例垂体腺瘤标本的MMP-9-PEX mRNA和MMP-9mRNA表达,免疫组化检测MMP-9蛋白表达;分析其与侵袭性垂体腺瘤的关系及两者之间的相关性。结果侵袭性垂体腺瘤组MMP-9-PEX mRNA表达较非侵袭性垂体腺瘤组降低(P<0.05);侵袭性垂体腺瘤组MMP-9的mRNA及蛋白表达均高于非侵袭性垂体腺瘤组(P<0.05);侵袭性垂体腺瘤组中MMP-9-PEX mRNA表达与MMP-9mRNA表达呈负相关(r=-0.66,P<0.05)。结论 MMP-9-PEX低表达及MMP-9高表达可能与垂体腺瘤的侵袭性生长密切相关;MMP-9-PEX及MMP-9可以作为垂体腺瘤侵袭性的参考指标,对诊断、治疗和预后判断有重要价值。更多还原

【Abstract】 Objective To investigate the relationship of matrix metalloproteinase 9(MMP-9)and MMP-9-PEX expressions and invasively growing of pituitary adenomas.Methods The expressions of MMP-9-PEX and MMP-9 mRNA were detected by RT-PCR,and the protein expression of MMP-9 was examined by immunohistochemistry.The correlation between MMP-9mRNA expression and MMP-9-PEX mRNA expression was analyzed.Results The mRNA expression of MMP-9-PEX was significantly lower in invasive pituitary adenomas than that in noninvasive pituitary adenomas(P<0.05).The expressions of MMP-9mRNA and protein were higher in invasive pituitary adenomas than those in noninvasive pituitary adenomas(P<0.05).The expression of MMP-9-PEX mRNA was negatively correlated to that of MMP-9 mRNA(r=-0.66,P<0.05).Conclusion The low expression of MMP-9-PEX and high expression of MMP-9 may be closely related to invasively growing of pituitary adenomas,which may be taken as the indicators for assessing the invasion of pituitary adenomas and have an important role in the diagnosis,treatment and prognosis evaluation of pituitary adenomas.更多还原
