力竭运动及恢复期间大鼠骨骼肌及心肌MSTN、TGF-β、PDGF含量的变化

【摘要】 目的:研究力竭运动及恢复期间大鼠骨骼肌及心肌肌肉生长抑制素(MSTN)、转化生长因子β(TGF-β)和血小板生长因子(PDGF)的变化。方法:Wistar雄性大鼠24只,随机分为4组,每组6只。E0组、E24组、E48组均进行一次性跑台力竭运动,并分别于力竭即刻、24小时后、48小时后处死;对照组(C组)不进行运动干预,随运动组后处死。取心肌与左侧腓肠肌,ELISA检测组织中MSTN、TGF-β和PDGF。结果:一次力竭运动后,与C组相比,E0组、E24组、E48组骨骼肌和心肌中MSTN、TGF-β的含量均显著性下降(P<0.05,P<0.01),PDGF含量则显著性升高(P<0.01);E0、E24、E48组间无显著性差异。结论:MSTN、TGF-β可能在骨骼肌、心肌修复及肌卫星细胞激活中起负调节作用,PDGF可能在骨骼肌、心肌修复中起正调节作用。

【Abstract】 Objective The paper investigated how exhaustive exercise affected the myostatin(MSTN),platelet growth factor(PDGF) and transforming growth factor-beta(TGF-β) in skeletal muscle and myocardium of rat. Methods 24 Wistar male rats were equally and randomly divided into group C(sedentary),group E0(sacrificed immediately after exhaustive exercise),group E24(sacrificed 24 hours after exhaustive exercise),and group E48(sacrificed 48 hours after exhaustive exercise). The gastrocnemius muscle and myocardium were removed for detecting MSTN, PDGF and TGF-β by ELISA. Result Compared with the group C,the contents of MSTN and TGF-β decreased significantly,and the content of PDGF increased significantly in skeletal muscle and myocardium after an exhaustive exercise(P<0.01)in groups E0, E24, and E48. There were no significant change in PDGF content among groups E0, E24, and E48. Conclusion MSTN and TGF-β could negatively adjust the muscle repair process and the activation of satellite cell,and PDGF positively promote the restoration of skeletal muscle and myocardium.
