生命早期不同营养状态对大鼠体质量及下丘脑mTOR表达的影响
Effect of different nutritional status during early life on the weight and hypothalamic m TOR expression in rats
【摘要】 目的研究生命早期的营养状态对各生长期大鼠体质量、下丘脑哺乳动物雷帕霉素靶蛋白(m TOR)表达的影响。方法将84只1日龄SD大鼠分为小窝组(40只)与对照组(44只,哺乳期死亡2只),母乳喂养至21日龄。断乳后将2组大鼠各自随机分为高脂组及普通组,于不同生长期取鼠测量体质量、Lee’s指数、内脏脂肪质量(VFM)及检测下丘脑m TOR的表达情况。结果未发育期小窝组体质量明显高于对照组(P<0.05)。自发育前期起,高脂喂养雌性大鼠Lee’s指数及VFM均高于普通喂养大鼠,高脂喂养的小窝大鼠的体质量明显高于普通喂养的对照大鼠(P均<0.05)。高脂喂养雄性大鼠体质量、Lee’s指数及VFM均高于普通喂养大鼠(P<0.05)。小窝组下丘脑m TOR的相对表达量高于对照组(P=0.015),高脂喂养大鼠下丘脑m TOR相对表达量高于普通喂养大鼠(P<0.05)。结论生命早期不同营养状态对大鼠不同生长时期的体质量有影响,并持续影响其下丘脑m TOR的表达,最终可能导致能量代谢紊乱。
【Abstract】 Objective To evaluate the effect of different nutritional status during early life upon the weight and the expression of hypothalamic mammalian target of rapamycin( m TOR) in rats of varying developmental stages. Methods Eighty-four Sprague-Dawley rats aged 1 day were assigned into the small( n = 40)and control groups( n = 44,2 rats died during lactation period),and breast-fed until 21 days of age. Then,the rats were randomly divided into the high-fat feeding and normal feeding groups. The rats’ weight,Lee’s index,visceral fat mass( VFM) and the expression of hypothalamic m TOR were measured at different developmental stages. Results The weight of prepubertal rats in the small group was significantly higher than that in the control group( P < 0. 05). Since early pubertal stage,the Lee’s index and VFM of female rats in the highfat feeding group were considerably higher than those in the normal feeding group( both P < 0. 05). The weight of rats fed with high-fat diet in the small group was significantly higher compared with that of control rats with normal feeding( P < 0. 05). The weight,Lee’s index and VFM of male rats with high-fat feeding were significantly higher compared with those of the control rats( all P < 0. 05). The relative expression of hypothalamic m TOR of the rats in the small group was dramatically higher than that of the animals in the control group( P =0. 015). The relative expression of hypothalamic m TOR in the rats with high-fat feeding was significantly higher than that in rats with normal feeding( P < 0. 05). Conclusions The nutritional status during early life exerts an effect upon the rat weight during different developmental stages and persistently affects the expression of hypothalamic m TOR,probably eventually leading to energy metabolism disorders.
