阿利吉仑对急性心肌梗死大鼠心肌间质重构和心功能的影响
Effects of Aliskiren on ventricular remodeling and heart function of rats with acute myocardial infarction
【摘要】 目的 探讨阿利吉仑对大鼠心肌梗死后左室重构和心功能的影响。方法 选取60只8周龄清洁级雄性Wistar大鼠,随机分为A组(假手术组)、B组(对照组)、C组(培哚普利组)和D组(阿利吉仑组),每组15只大鼠。B、C、D组大鼠建立心肌梗死模型,A组只将缝线绕过冠状动脉但不予结扎。术后C、D组分别给予培哚普利(2.5 mg·kg-1·d-1)和阿利吉仑(50 mg·kg-1·d-1)灌胃,共持续8周。实验过程中进行以下检测:开始及结束时进行超声心动图检查,测量左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVDs)、舒张末期室间隔厚度(IVSd)、舒张末期左心室后壁厚度(LVPWd)、左心室射血分数(EF)、左心室短轴缩短率(FS)及二尖瓣瓣口舒张早期、晚期最大血流峰值速度(E峰、A峰)和E/A。处死动物后测量心脏重量、左心室重量,计算心脏重量指数、左心室重量指数。结果 ①B组心脏重量指数、左心室重量指数、LVEDD、LVDs较A组显著增高(P<0.05);而1VSd、LVPWd、LVEF、FS、E/A显著低于A组(P<0.05);与B组比,C和D组心脏重量指数、左心室重量指数、LVEDD、LVDs显著降低(P<0.05),而IVSd、LVPWd、LVEF、FS、E/A显著高于B组(P<0.05)。②B、C、D组大鼠心肌胶原容积分数(10.5±1.3、6.5±0.7、6.7±0.6)较A组明显升高(P<0.05);而C、D组较B组心肌胶原容积分数显著降低(P<0.05);C组和D组之间比较无明显差别(P>0.05)。结论 ①阿利吉仑干预能减轻大鼠心肌梗死后左室重构,改善心功能。②阿利吉仑和培哚普利在减轻大鼠心肌梗死后左室重构、改善心功能方面无明显差别。
【Abstract】 Objective The aim of the study was to investigate the effect of Aliskiren on ventricular remodeling and heart function in the rats with acute myocardial infarction.Methods 60 male 8-week-old Wistar rats were enrolled.After one week,they were randomly divided into four groups(n—5,per group):sham operated group,control group,Perindopril group and Aliskiren group.The rats in control group,Perindopril group and Aliskiren group were all ligated the anterior descending coronary branch for developing an animal model of anterior myocardial infarction.Meanwhile,sham operated group was regarded as a concurrent group and the rats were subjected to sham coronary ligature.The next day of the operation,the rats in Perindopril group and Aliskiren group were dosed orally per day with Perindopril(2.5 mg·kg-1·d-1) or Aliskiren(50 mg·kg-1·d-1) for 8 weeks,respectively.At the same time,the animals in sham operated group,control group were provided with 0.9%saline.We deteced the parameters as follows:(1)Before at the end of the experiment,all the rats were weighed and the pressure blood were assayed.(2)Echocardiographic images were acquired before and at the end of the experiment.The left ventricular internal diameter end-diastole(LVEDD),left ventricular internal diameter end-sy9tole(LVDs),interventricular septum thickness at enddiastole(IVSd),left ventricle posterior wall at enddiastole(LVPWd),left ventricle ejection fration(LVEF),fractional shortening(FS),the early diastolic(E),late diastolic(A) velocities and E/A of atrioventricular valve were measured.(3)The rats were killed and the samples were obtained.The weight of heart and left ventricle were measured,cardiac mass index(CMI) and left ventricular mass index(LVMI) were calculated.Results(I)The rats in sham operated group displayed normal echocardiographic images.The rats in control group showed the worsened of left ventricular.Compared to sham operated group,the values of CMI,LVMI,LVEDD and LVDs were significantly increased,and IVSd,LVPWd,LVEF,FS,E/A were significantly decreased(P<0.05).In comparision with control group,CMI,LVMI,LVEDD and LVDs were significantly decreased,and IVSd,LVPWd,LVEF,FS,E/A were significandy increased(P<0.05).So,the disfunction was reversed in Perindopril group and Aliskiren group.(2)The value of CVF was higher in control group,Perindopril group and Aliskiren group(10.5±1.3,6.5±0.7,6.7±0.6) than that in sham operated group(P<0.05).The value of CVF was lower in Perindopril group and Aliskiren group than that in control group(P<0.05).There was no difference between Perindopril group and Aliskiren group(P>0.05).Conclusion(l)Aliskiren had a protective effect on ventricular remodeling and cardiac function in the rats with AMI.(2) There was no difference of Perindopril and Aliskiren treatment on the beneficial effect of ventricular remodeling and heart function in rats of AMI.
