荸荠种质资源品种遗传多样性ISSR分析

【摘要】 为揭示不同荸荠栽培品种的遗传多样性,利用ISSR分子标记技术对24个荸荠品种的亲缘关系进行分析。结果表明,从100条ISSR引物中筛选出适合荸荠种质分析的11条ISSR引物,共扩增出86条带,平均每条引物扩增出7.8条带,多态性为69条带,多态性比例为80.3%。供试的24份荸荠品种两两之间的遗传距离值在0.0909-0.6071之间,平均值为0.3493。UPGMA法聚类结果表明24个荸荠品种在遗传距离0.34处可分为五类。24个荸荠品种在ISSR分析中亲缘关系较近,其遗传基础相对较狭窄,但部分不同地区栽培品种之间仍存在一定的遗传差异。

【Abstract】 The paper focused on assess the genetic variability and diversity of water chestnut( Eleocharis tuberosa Schulut). Genetic relation among 24 cultivars of water chestnut was analyzed by inter-simple sequence repeat( ISSR) makers. 11 polymorphism primers were selected from 100 ISSR primers. A total of 86 ISSR bands were obtained,that is,7. 8 polymorphic ban each primer pair,and 69 of them were polymorphic,the polymorphism ratio was 80. 3 %. The dissimilarity coefficient of the 24 water chestnut cultivars ranged from 0. 0909 to 0. 6071,with an average of 0. 3493. The 24 water chestnut cultivars were divided into 5 groups at the genetic distance 0. 34 by UPGMA. The results showed that narrow genetic diversity and close genetic relationship presented among 24 water chestnut cultivars,and the difference varied in part of those cultivars from different areas,and the same as RAPD analysis at prophase.
