过表达HIF-1α调控HO-1对糖尿病大鼠心肌缺血再灌注损伤作用的研究

【摘要】 目的探讨过表达缺氧诱导因子-1α(HIF-1α)对糖尿病心肌缺血再灌注(I/R)损伤的影响,并对其作用机制做初步探讨。方法构建重组HIF-1α真核表达载体,用链脲佐菌素(STZ)诱导建立糖尿病大鼠模型,动物分为正常假手术组(C组)、缺血再灌注组(I/R组)和I/R加HIF-1α诱导组(I/R+HIF-1α组),每组12只大鼠,共36只。检测各组大鼠心肌病理损伤分级、心肌坏死面积、HIF-1α和HO-1的蛋白表达。结果 I/R+HIF-1α组心肌病理损伤分级及心肌坏死面积与I/R组相比明显减少(P<0.05),I/R+HIF-1α组心肌HIF-1α的蛋白表达与I/R组相比明显增加(P<0.05),I/R+HIF-1α组心肌HO-1的蛋白表达与I/R组相比明显增加(P<0.05)。结论过表达HIF-1α,可以调控HO-1的表达,减轻糖尿病大鼠心肌的缺血再灌注损伤,对防治糖尿病大鼠心肌缺血再灌注损伤有一定的作用。

【Abstract】 Objective To explore the role of over-expression of hypoxia inducible factor 1α on myocardial ischemia reperfusion injury and its potential mechanisms. Methods The recombinant mammal expression vector encoding rat hypoxia inducible factor 1α was conducted. The diabetic rats were confirmed by intraperitoneal injection of streptozotocin(STZ, 50mg/kg). The Wister rats randomly divided into three groups: a sham group(C group, 12 cases), an ischemia/reperfusion group(I/R group, 12 cases), and an I/R plus HIF-1α group(I/R+HIF-1αgroup, 12 cases). At the end of the study,the infarct size was determined by HE staining. The protein expressions of HIF-1α and HO-1 were detected by Western blot. Results the over-expression of HIF-1α reduced myocardial ischemia/reperfusion injury as evidenced by the significant decrease of myocardial infarct size(P< 0.05). Furthermore, the protein expressions of HIF-1α and HO-1 markedly were increased in the I/R+HIF-1α group compared with those in the I/R group(P<0.05). Conclusions Over-expression of HIF-1α has induced the notable increase of HO-1 and has effects on anti-myocardial ischemia/reperfusion in the diabetic rats.
