从放人员染色体、微核检查的制片方法改进探讨
Discussion on improved methodology for chromosome and micronucleus health examination for radiation related workers
【摘要】 目的对提高放射工作人员健康检查中染色体检查和微核检查效率进行实验方法的探讨。方法通过对人类外周血染色体制片和微核制片的收获过程进行改进,将两种标本制片在同一张玻片上。在染色体标本低渗之后,与微核标本混合,使实验中预固定、固定、滴片和染色步骤合并,从而实现同一张玻片上既包括染色体畸变标本,又包括微核标本。结果采集正常标本10份,受辐照标本10份。对于同一个体标本,采用改进方法检测到的染色体畸变率和微核率结果与分别制作染色体片和微核片检测的结果一致。结论该方法使该项工作在一定程度上得到简化,在同样实现检测效果的前提下,既节省了成本,又提高了工作效率。
【Abstract】 Objective The aim of this study was to discuss the efficiency of different methodology of chromosome and micronucleus health examination for radiation related workers. Methods Instead of making separate chromosomes and micronucleus specimens,the chromosome specimen after hypotonic was mixed with micronuclei specimen. Then the processes of pre-fix, fix, drop sheet and dying were combined, so the observation of chromosome aberration and micronucleus rate was done on the same slide. Results With ten specimens collected from irradiated samples and ten specimens from un-irradiated samples, the results of chromosome aberration and micronucleus rate observed on the same slide were consistent with the results from separate slides for the same individual. Conclusion This new methodology allowed the work to be simplified to a certain extent to achieve the same level of detection effect, while saving costs and improving efficiency.
