老鼠簕醇提物抗鸭乙型肝炎病毒及护肝作用的实验研究
The anti-viral effect against duck hepatitis B and liver-protection function of the alcohol extract of Acanthus ilicifolius L
【摘要】 目的观察老鼠簕醇提物(Elcohol Extract of Acanthus ilicifolius L.,EEA)治疗鸭乙型肝炎的疗效。方法将60只感染鸭乙型肝炎病毒(DHBV)的麻鸭随机分为5组:EEA低、中、高剂量组、阿昔洛韦(ACV)组和模型对照组。另设1组正常对照组。每组12只,连续给药14 d。用FQ-PCR法检测治疗前(T0)、用药后d 7(T7)、d 14(T14)及停止治疗后d 5(P5)血清中DHBV DNA的水平,ELISA法测定血清中DHBs Ag、DHBe Ag的OD值,同时检测血清AST、ALT,最后取肝脏做病理检查。结果老鼠簕(Acanthus Ilicifolius L.,AIL)各剂量组血清DHBV DNA水平、DHBs Ag及DHBe Ag OD值均未显著降低(P>0.05)。高、中剂量EEA能显著降低ALT活性(P<0.05);高剂量EEA显著降低AST活性(P<0.01)及显著改善肝脏病理作用(P<0.001)。结论高剂量EEA对肝功能及肝组织有明显的保护作用,但未发现EEA有抗鸭乙肝病毒作用。
【Abstract】 Objective This work was to evaluate the efficacy of alcohol extract of Acanthus ilicifolius L.(EEA) on duck hepatitis B.Methods 60 ducks infected with DHBV were randomly divided into low, middle and high dosage groups of EEA, acyclovir(ACV)group and model control group. Another normal control group was also included in the study by using non-DHBV infected ducks.Each group had 12 ducks and drugs were given for 14 days. The levels of serum DHBV DNA were detected by fluorescence quantitative PCR(FQ-PCR) before treatment(T0), 7 days(T7) and 14 days(T14) after drug administration, 5 days after treatment(P5). ELISA was used to measure the OD values of DHBs Ag and DHBe Ag in serum. ALT and AST in serum were measured at the same time. Finally livers were taken for histopathological examination. Results The levels of serum DHBV DNA, the ODs of DHBs Ag and DHBe Ag were not significantly reduced among all dosage groups(P>0.05). However, the ALT activity was obviously lower in the middle and high dosage groups(P<0.05). It was also found that high dosage alcohol extract could significantly reduce the activity of AST(P<0.01) and improve hepatic pathological effect(P<0.001). Conclusion High-dosage EEA has an obvious protective effect on the liver function and liver tissue, but had no antiviral effect against duck hepatitis B.
