镉损伤小鼠肾脏形态学研究及姜黄素的保护作用

【摘要】 采用腹腔注射CdCl2(2mg/kg/day)和灌胃不同浓度的姜黄素,连续处理7天构建染镉小鼠模型,通过丙二醛(MDA)含量测试小鼠体内脂质过氧化水平验证模型是否构建成功,利用光镜技术观察镉致肾脏组织形态学的改变,通过元素分析以及金属螯合实验探讨姜黄素保护镉致肾脏损伤的保护机制.结果显示,镉的毒性能够引起肾脏损伤组空泡增多并出现细胞炎症侵润,细胞核发生聚集现象,而姜黄素保护可以减少空泡的形成;同时元素分析表明镉可以在肾脏中蓄积,影响必需元素的吸收,而姜黄素可以呈剂量依赖螯合Cd2+,进而抑制镉对肾脏的损伤.

【Abstract】 This research was designed to study the effect of curcumin on cadmium induced mice renal damage.Mice was treated by intraperitoneal injection of CdCl2(2mg/kg/day)and lavage different concentrations of curcumin for 7days.The content of MDA was used to test lipid peroxide(LPO)in mice and detect whether the model was built successfully,the renal histopathological changes induced by cadmium were observed using light microscope and the action mechanism of curcumin against the toxicity of Cd was explored by elemental content analysis and metal chelating assay.Results show that the intoxication of Cd can lead to the increase of vacuolus,the infiltration of inflammatory cells and the swelling of myocytes in damage group,however,the protective effect of curcumin can reduce the occurrence of histopathological changes.Meantime,element analysis results showed that the accumulation of cadmium in kidneys can,affect the absorption of essential elements,and curcumin show doseresponse ability of chelating Cd2+,thereby inhibiting cadmium damage to kidneys.
