【摘要】 目的探讨血脂康胶囊对脂多糖(LPS)诱导大鼠肝脏Kupffer细胞NF-κB表达的影响。方法将分离的Kupffer细胞按1×105个/ml接种于24孔板内培养48 h后,对照组:常规培养;LPS组:给予终浓度为100 ng/ml的LPS培养8 h;血脂康胶囊组:血脂康胶囊用生理盐水溶解,90 mg·kg-1·d-1,其余处理同LPS组。ELISA法检测上清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)水平。Western blotting检测胞浆IκBα、p-IκBα及胞核NF-κB p65蛋白表达水平。结果 LPS组细胞上清TNF-α、IL-1β和IL-6表达高于对照组(P<0.05)。血脂康胶囊组细胞上清TNF-α、IL-1β和IL-6低于LPS组(P<0.05),但高于对照组(P<0.05)。对照组胞浆蛋白IκBα表达显著高于LPS组和血脂康胶囊组(P<0.05),但LPS组明显低于血脂康胶囊组(P<0.05)。LPS组胞核NF-κB p65和p-IκBα表达高于对照组和血脂康胶囊组(P<0.05),但血脂康胶囊组表达低于LPS组(P<0.05)。结论血脂康胶囊对LPS诱导的Kupffer细胞炎症因子分泌有一定的抑制作用。更多还原

【Abstract】 Objective To investigate the effect of Fat Kang Capsule on the expression of NF-κB in Kupffer cells of cells induced by lipopolysaccharide( LPS). Methods The Kupffer cells were inoculated into 24 hole plate for 48 hours,control group: routine culture; LPS group: culture with LPS for 8 hours; Fat Kang Capsule group: culturel with physiological Saline. The change of TNF-α,IL-1β and IL-6 were detected by ELISA. The level of IκBα,p-IκBα,NF-κB p65 were detected by Western blotting. Results The expression of IL-6,IL-1β and TNF-α in LPS group were higher than those in control group( P < 0. 05). IL-6,IL-1β and TNF-α of cell supernatant of Fat Kang Capsule were lower than those in LPS group( P < 0. 05),while higher than those in control group( P < 0. 05). The expression of cytosolic protein IκBα in control group was higher than that in LPS group and Fat Kang Capsule group( P < 0. 05). While it in LPS group was lower than that of the Fat Kang Capsule group( P < 0. 05). The expression of NF-κB p65,p-IκBα in LPS group were higher than those in control group( P < 0. 05). While those in Fat Kang Capsule group were lower than those in LPS group( P < 0. 05). Conclusion Fat Kang Capsule has a certain inhibition effect on the inflammatory factors of the Kupffer cells induced by LPS are secreted.更多还原
