【摘要】 探讨化瘀通络中药对糖尿病肾病(DN)大鼠的干预作用和对肾组织纤维连接蛋白(FN)和层粘连蛋白(LN)的影响。选用SD雄性大鼠55只,随机取10只作为正常对照组(C组),其余利用高脂饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射建立糖尿病(DM)大鼠模型,成模大鼠分组如下:模型组(M组)、化瘀中药治疗组(H组)、通络中药治疗组(T组)、厄贝沙坦治疗组(I组)。各治疗组给予相应药物灌胃16周,于16周末检测大鼠空腹血糖(FBG)、24 h尿蛋白定量(24 h UTP)、凝血酶原时间(PT)、血肌酐(Scr)、甘油三脂(TG);取大鼠肾组织做病理学观察;免疫组化法检测肾组织FN和LN的表达。结果显示与正常对照组比较,模型组大鼠FBG、24h UTP、PT、TG、Scr水平明显升高(P<0.05),FN、LN表达明显增加(P<0.05)。与模型组比较,各治疗组大鼠24h UTP、PT、TG水平明显降低(P<0.05),FN、LN表达明显减少(P<0.05)。以上结果说明化瘀通络中药对糖尿病肾病大鼠的肾脏有一定的保护作用,其机制可能与调节脂代谢紊乱,纠正血液高凝状态,下调FN、LN表达,延缓肾脏纤维化有关。更多还原

【Abstract】 To explore the intervention effect of Huayu Tongluo traditional Chinese medicine on diabetic nephropathy( DN) rats and its effect on kidney fibronectin and laminin expression.Ten rats were randomly selected as normal control group( C group) from 55 male Sprague-Dauley rats,and the remaining rats were established by high-fat diet combined with low-dose streptozotocin intraperitoneal injection.Successful model rats were randomly divided into four groups,namely: model group( M group),Huayu recipe treatment group(H group),Tuolong recipe treatment group( T group),Irbesartan group( I group).Each treatment group gave corresponding drug lavage,once daily for 16 weeks.At the end of the 16 thweek,The FBG,24 h UTP,PT,Scr,TG were tested.The renal tissues of rats were separated for pathological observation.The expression of FN and LN in renal tissue was tested by immunohistochemistry.Compared with control group,the FBG,24 h UTP,PT,TG and Scr levels of model group were significantly increased( P < 0.05),the expression of FN and LN were significantly increased( P < 0.05); compared with model group,the 24 h UTP,PT,TG levels of every treatment group were significantly reduced( P < 0.05),the expression of FN and LN were significantly reduced( P <0.05).These results suggested that Huayu Tongluo traditional Chinese medicine showed a protective effect in the DN rats kidney,suggesting its therapeutic effect on DN rats may be related to the regulation of lipid metabolism,correction of blood hypercoagulable state,reduction of FN and LN expression and delay renal fibrosis.更多还原
