敦煌固本方对运动性疲劳小鼠心肌组织能量代谢酶及超微结构影响的实验研究

【摘要】 目的:探讨敦煌固本方对运动性疲劳小鼠心肌组织能量代谢酶及超微结构的作用。方法:90只小鼠,随机分为6组,分别为安静对照组(C)、安静给药组(E)、运动对照组(TC)、运动给药组(TE),运动组又各分为力竭运动后即刻组和力竭运动后恢复24小时组,即TC0、TC24、TE0、TE24。给药组每天以20/mg/(kg·d)灌服敦煌固本方,对照组灌同体积的生理盐水。运动组进行6周的递增强度游泳训练,然后进行最后一次无负重的力竭性游泳,形成慢性力竭性运动性疲劳。运动后即刻组在力竭运动后即刻处死,运动后恢复24小时组力竭运动后恢复24小时后处死,处死后立刻取出各组心肌组织,测定心肌组织能量代谢酶(CK、LDH、SDH)及Na+,K+_ATP酶的活性;用透射电镜观察心肌组织超微结构的改变。结果:1)安静时,E组与C组比较,小鼠心肌组织CK、LDH、SDH活性和Na+,K+-ATP酶活力无明显变化(P>0.05)。运动后即刻,小鼠心肌组织CK、LDH、SDH活性下降,TC0和TE0组显著低于C组(P<0.05),但TE0组明显高于TC0组,差异有显著性(P<0.05);与C组比较,TC0组小鼠心肌组织Na+,K+_ATP酶活力略有下降,但无显著性差异(P>0.05);TE0组小鼠心肌组织Na+,K+_ATP酶显著高于TC0组(P<0.05)。运动后恢复24小时组,小鼠心肌组织CK、LDH、SDH活性均有所恢复,但是仍然没有恢复到安静时的水平,但是,TE24组小鼠心肌组织CK、LDH、SDH活性显著高于TC24组(P<0.05);与TC0组相比,TC24组Na+,K+_ATP酶活力略有升高,但无显著性差异(P>0.05),但TE24组小鼠心肌组织Na+,K+_ATP酶活力显著高于TC24组(P<0.05)。2)力竭运动可引起心肌组织超微结构的损伤,而灌服敦煌固本方小鼠心肌组织超微结构损伤与对照组相比明显减轻。结论:敦煌固本方可以改善运动性疲劳小鼠的心肌能量供应,提高小鼠运动能力,对力竭运动后心肌损伤具有明显的保护作用。

【Abstract】 Objective: To evaluate the effects of Dunhuang Guben Fang on myocardial energy metabolism enzymes and ultrastructure changes in mice induced by exercise fatigue. Methods: 90 Kunmin male mice were randomly divided into six groups: rest control group( C),rest experiment group( E),exercise training control group( TC),exercise training experiment group( TE). The experiment group was administered Dunhuang Guben Fang once a day,rest control group,exercise training control group were fed an equal volume of NS,training groups carried out a 6- week swimming program with gradually increasing intensity and an exhaustive swimming before sacrifice,the trained mice were sacrificed in four kinds of conditions: immediately after exhaustive exercise in control group( TC0),immediately after exhaustive exercise in exercise training experiment group( TE0),24-hour’s recovery after exhaustive exercise in control group( TC24),24- hour’s recovery after exhaustive exercise in exercise training experiment group( TE24). The changes of CK、LDH、SDH and Na+,K+- ATP ase activitiesof myocardium in mice were determined,the ultrastructure changes of myocardium were observed by transmission electron microscope( TEM). Results: 1) There was no remarkable difference between the group C and E in the activities of CK,LDH,SDH and Na+,K+- ATP ase at reststates( P > 0. 05). In immediately after exhaustive exercise,the activities of CK,LDH,SDH were obvious lower than those of control group( P < 0. 05). Compared with group C,the activities of Na+,K+_ATP ase wasn’t significant( P > 0. 05). But those were higher in experiment groups than those in control groups( P < 0. 05). After 24-hour recovery,CK,LDH,SDH and Na+,K+_ATP ase have significant difference between experiment group and its control group( P < 0. 05). 2) Ultrastructure observation showed that after exhaustive swimming the cardiac muscle lesion of Dunhuang Guben Fang group was less severe than in control group. Conclusions: It was proved that Dunhuang Guben Fang could improve the rats’ energy metabolism,delay the sports fatigue,and effectively protect cardiac muscle against free radical damage.
