互隔交链孢霉素对NIH/3T3细胞的基因损伤及DNA聚合酶β表达的影响
The genetic damage of altenuene alternate to NIH / 3T3 cells and its effects on the expression of DNA polymerase β
【摘要】 目的:研究互隔交链孢霉素(altenuene,ALT)对NIH/3T3细胞的基因损伤作用及对DNA聚合酶β(DNA polymeraseβ,DNApolβ)表达的影响,探讨互隔交链孢霉(Alternaria alternate)致癌机制。方法 :用单细胞凝胶电泳检测不同浓度的ALT对NIH/3T3细胞的基因损伤作用;以半定量RT-PCR及Western-blot方法分别检测DNApolβ在基因及蛋白水平的表达。结果:10.0、20.0、50.0μmol/L的ALT作用于NIH/3T3细胞后,细胞尾长及尾部DNA含量均增加,一定浓度的ALT作用于细胞后DNApolβ在基因及蛋白水平表达均上调,与溶剂对照组相比,差异均有统计学意义(P<0.05)。结论:ALT可引起NIH/3T3细胞基因损伤,且有剂量依赖性,并可导致DNApolβ的表达上调。
【Abstract】 Objective To study the effect on genetic damage of altenuene(ALT) and the expression of DNA polymerase β(CDNApo1β) in NIH / 3T3 cells, and discuss the carcinogenesis mechanism of ALT. Methods Single cell gel electrophoresis was used to measure genetic damage to NIH / 3T3 cells. RT-PCR and Western-blot were used to detect the expressions of DNApolβ on m RNA and protein level respectively. Results The tail length and content of tail DNA were all increased after treatment with ALT of 10.0, 20.0, 50.0 μmol / L, and the difference was statistically significant(P <0.05). The expression of DNApolβ was increased at both the m RNA and protein level after treatment by 20.0 μmol / L ALT. Conclusion ALT may cause gene damage even up to doses and induce DNApolβ overexpression in NIH / 3T3 cells.
