【摘要】 目的建立猪链球菌2型感染胶体金快速诊断方法。方法选择猪链球菌在进化上极为保守的蛋白谷氨酸脱氢酶的编码基因gdh作为研究靶基因,对其进行克隆、表达和纯化,应用硝酸纤维膜包被纯化的目的蛋白,运用胶体金溶液与生物大分子金黄色葡萄球菌A蛋白(SPA)结合制备SPA金标探针建立猪链球菌2型感染的胶体金快速诊断方法。结果胶体金检测标本阳性结果试纸条上出现两条红色条带,阴性结果试纸条上只有一条靠近吸水端的质控线出现红色条带,对370份标本进行检测,其中320份阴性结果,50份阳性结果。结论运用胶体金免疫层析技术可以快速诊断猪链球菌2型感染。更多还原

【Abstract】 Objective To establish a fast colloidal gold diagnostic assay for Streptococcus suis serotype 2 infection. Methods The conservative protein glutamate dehydrogenase( GDH) encoding gene gdh was selected as the target gene for cloning,expression and purification. A rapid colloidal gold diagnostic assay for Streptococcus suis serotype 2 infection was established by using microporous membrane carrier such as nitrocellulose membrane to envelop the purified protein and by using gold granule to label Staphylococcus aureus protein A( SPA). Results Two red bands appeared in the positive specimens detected by colloidal gold test strip,while only one red band appeared on a quality control line strip near the suction end in the negative specimens. A total of 370 specimens were detected,and 320 were negative and 50 were positive. Conclusions Applying colloidal gold immunochromatography assay can rapidly diagnose Streptococcus suis serotype 2 infection.更多还原
