3种金黄色葡萄球菌基因分型技术的评估

【摘要】 目的应用3种金葡菌基因分型方法分析食物中毒来源金葡菌,综合评价3种方法在金葡菌监测或暴发调查的可行性。方法应用多位点熔解曲线体系(MLMC)、葡萄球菌蛋白A序列分析(Spa typing)及脉冲场凝胶电泳(PFGE)三种基因分型技术对59株金葡菌进行分子分型。结果多位点熔解曲线分型体系将59株金葡菌分为19个型别,PFGE将其分为15个型别,Spa typing分为10个型别。Simposn差异指数分别是0.92、0.87和0.85。结论 3种分型方法应用于12起食物中毒调查,均能在同一起疫情中分析到型别一致的菌株,证实了菌株之间的亲缘关系。MLMC简单、快速,分辨率高于PFGE和Spa typing分型技术,适用于同一批暴发菌株的初筛。合理使用熔解曲线、Spa typing及PFGE分型方法对致病性金葡菌的防治具有重要意义。

【Abstract】 Objective To analyze the genotypes of Staphylococcus aureus strains isolated from the suspected food- borne diseases using 3 different genotyping methods,and to comprehensively evaluate the feasibility of the 3 genotyping methods for monitoring Staphylococcus aureus and survey of the outbreak. Methods Fifty- nine strains of Staphylococcus aureus were analyzed on the basis of multiple locus melting curve system( MLMC),Staphylococcus aureus protein A( Spa) typing and pulsed field gel electrophoresis( PFGE). Results Fifty- nine strains were classified into 19 genotypes by MLMC,15 genotypes by PFGE and 10 genotypes by Spa typing. The Simpson’s diversity indices for MLMC,PFGE and Spa typing were 0. 92,0. 87 and 0. 85 respectively. Conclusions Twelve food poisoning outbreaks detected by the 3 genotyping methods indicate that the strains with the same genotype are found in the same outbreaks,demonstrating the genetic association of the strains. The discriminatory power of MLMC is higher than that of PFGE and Spa typing,and as a simple and fast genotyping method,it could be used as a first- line typing tool for analyzing outbreaks of Staphylococcus aureus. Rational combination of these 3 tools will be of great significance in pathogenic Staphylococcus aureus control and prevention.
