一氧化碳释放分子对病毒性心肌炎小鼠的保护作用及其作用机制研究
Protective Effect of Carbon Monoxide Releasing Molecule on Viral Myocarditis Mice and Its Mechanisms
【摘要】 目的探讨一氧化碳释放分子(CORM)对病毒性心肌炎(VMC)小鼠的保护作用及其作用机制。方法将45只4周龄雄性纯种BALB/C小鼠随机分为正常对照组、空白对照组及CORM干预组,每组15只,制备VMC模型,CORM干预组小鼠予以3 mg/ml CORM溶液0.1 ml腹腔注射,正常对照组和空白对照组小鼠予以二甲基亚砜0.1ml腹腔注射,记录各组小鼠死亡数量,检测各组小鼠血清心肌肌钙蛋白I(c Tn I)浓度及心肌匀浆液丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、IL-1β、IL-6、IL-18、肿瘤坏死因子α(TNF-α)浓度,计算各组小鼠心肌病变积分。结果正常对照组、CORM干预组小鼠病死率均低于空白对照组(P<0.05);空白对照组血清c Tn I浓度、心肌病变积分高于正常对照组和CORM干预组,CORM干预组血清c Tn I浓度、心肌病变积分高于正常对照组(P<0.05)。空白对照组心肌匀浆液MDA浓度高于正常对照组和CORM干预组,心肌匀浆液SOD、GSH-Px、CAT浓度低于正常对照组和CORM干预组,而CORM干预组心肌匀浆液MDA浓度高于正常对照组,心肌匀浆液SOD、GSH-Px、CAT浓度低于正常对照组(P<0.05)。空白对照组心肌匀浆液IL-1β、IL-6、IL-18、TNF-α浓度高于正常对照组和CORM干预组,CORM干预组心肌匀浆液IL-1β、IL-6、IL-18、TNF-α浓度高于正常对照组(P<0.05)。结论 CORM可降低VMC小鼠病死率及心肌损伤,具有一定的保护作用,其作用机制可能包括抑制脂质过氧化、减轻炎性反应等。
【Abstract】 Objective To investigate the protective effect of carbon monoxide releasing molecule( CORM) on viral myocarditis( VMC) mice,and explore its mechanisms. Methods Fifty- five 4- week old male BALB / C mice were randomly divided into normal control group, blank control group and CORM intervention group, each of 15 mice. VMC experimental models were made,and then CORM intervention group given CORM intraperitoneal injection( 0. 1 ml),normal control group and blank control group given DMSO intraperitoneal injection( 0. 1 ml). Incidence of death was recorded, serum of cardiac troponin I( c Tn I) level and myocardial homogenate levels of malondialdehyde( MDA), superoxide dismutase( SOD),glutathione- peroxidase( GSH- Px), catalase( CAT), IL- 1β, IL- 6, IL- 18, tumor necrosis factor- α( TNF- α)were detected,myocardial lesions score was calculated. Results The mortality of normal control group and CORM intervention group was lower than that of blank control group,respectively( P < 0. 05). Serum c Tn I level,myocardial lesions score of blank control group were higher than those of normal control group and CORM intervention group, and those of CORM intervention group were higher than those of normal control group( P < 0. 05). Myocardial homogenate MDA level of blank control group was higher than that of normal control group and CORM intervention group,respectively,while SOD,GSH- Px,CAT were lower( P < 0. 05); myocardial homogenate MDA level of CORM intervention group was higher than that of normal control group,while SOD,GSH- Px,CAT were lower( P < 0. 05). Myocardial homogenate levels of IL- 1β,IL- 6,IL- 18,TNF- α of blank control group were higher than that of normal control group and CORM intervention group, respectively, and those index of CORM intervention group were higher than those of normal control group( P < 0. 05). Conclusion CORM can reduce the mortality and myocardial damage of VMC mice, has certain protective effect; its possible mechanisms including control lipid peroxidation and reduce inflammatory reaction.
