大蒜素对NMBA诱发食管癌大鼠免疫功能的影响
The effect of allicin on immunologic function in rats with NMBA-induced esophagus cancer
【摘要】 目的研究大蒜素对甲基苄基亚硝胺(NMBA)诱发食管癌大鼠免疫功能的影响。方法 90只SD大鼠随机分成三组,每组30只。Ⅰ组:常规饲养,无任何处理;Ⅱ组:每周一次NMBA皮下注射,10 mg/kg;Ⅲ组:每周一次NMBA皮下注射,10 mg/kg。同时给予大蒜素15 mg/(kg·d),肌注给药。连续22周后,流式细胞仪测定大鼠外周血T淋巴细胞CD3+、CD4+、CD8+亚型及CD4+/CD8+。处死实验鼠并测量脾重系数。结果三组大鼠脾重量系数差异无统计学意义(P>0.05)。Ⅰ组实验鼠外周血T淋巴细胞亚群CD8+下降,而CD3+、CD4+、CD4+/CD8+均明显高于其他两组(P<0.05)。Ⅲ组CD3+、CD4+、CD4+/CD8+较Ⅱ组增高,而CD8+则低于Ⅱ组(P<0.05),差异有统计学意义。结论大蒜素对NMBA诱发的大鼠食管癌有明显的抑制作用,且能够明显增强食管癌大鼠的免疫系统功能。
【Abstract】 Objective To investigate the effect of allicin on immunologic function in rats with N-Nitrosomethylbenzylamine(NMBA)-induced esophagus cancer. Methods Ninety SD rats were divided into three groups randomly with 30 rats in each group: no treatment in group A,10 mg/kg of NMBA subcutaneous injection once a week in group B,10 mg/kg of NMBA subcutaneous injection once a week and 15 mg/(kg·d) of allicin intramuscular injection in group C. After 22 weeks of treatment,the peripheral blood T lymphocyte subtype CD3+,CD4+,CD8+and CD4+/CD8+ratio were analyzed with flow cytometry and the rats spleen weight coefficient was determined. Results The differences in spleen weight coefficients among the three groups were not statistically significant(P > 0. 05). The CD3+,CD4+,CD4+/CD8+were lower,but CD8+was higher in group B and group C than those in group A(P < 0. 05). The CD3+,CD4+,CD4+/CD8+of the group C were higher than group B,but CD8+was lower than group B(P < 0. 05). The differences were statistically significant. Conclusion Allicin can significantly enhance immunologic function of the rats with NMBA-induced esophageal cancer.
