荧光光谱法研究纳米TiO2与肌红蛋白的相互作用
Study on interaction of myoglobin with TiO2 nanoparticles by fluorescence spectroscopy
【摘要】 采用溶胶-凝胶法合成TiO2纳米颗粒,通过TEM对纳米TiO2进行了表征。结果表明,纳米TiO2的粒径约为56 mm。利用荧光光谱法研究了纳米TiO2与肌红蛋白(Mb)的相互作用,研究表明这种相互作用能使肌红蛋白的内源荧光猝灭。通过猝灭常数、结合常数和结合位点数的计算,证明此猝灭为静态猝灭机制,结合位点数约为1。根据热力学参数确定了它们之间的作用力主要是静电力和疏水力,使肌红蛋白构象发生了变化。
【Abstract】 TiO2 nanoparticles is synthesized by using the Sol-gel method.The morphology of TiO2 nanoparticles are characterized by the transmission electron microscopy(TEM).The interaction between TiO2 and myoglobin(Mb) is studied by using the fluorescence spectroscopic method.TiO2 effectively quenches the intrinsic fluorescence of Mb via static quenching.The process of binding TiO2 on Mb is a spontaneous molecular interaction procedure.According to thermodynamic parameters,△H and AS,it is indicated that the hydrophobic and electrostatic interactions can play a major role in stabilizing the TiO2-Mb complex.
