胶质母细胞瘤患者血浆miRNA表达谱研究

【摘要】 目的采用microRNA(miRNA)基因芯片分析在胶质母细胞瘤(GBM)患者血浆中表达的miRNA,寻找GBM血液中的分子生物学标志物。方法分别收集22例GBM患者和8例健康志愿者血液标本,试剂盒提取总RNA,与基因芯片进行杂交,检测GBM患者和健康志愿者血浆中miRNA表达谱,筛选差异表达的miRNA。采用Kaplan-Meier方法分析差异表达的miRNA与GBM生存期的关系。结果 miRNA表达谱分析发现在GBM差异表达的miRNA共19个,其中上调3个,下调16个。生存分析显示,miR-30c-2-3p高表达组患者中位无疾病进展时间较低表达组明显缩短(P=0.016)。结论通过基因芯片的方法发现了在GBM患者血浆中19个差异表达的miRNA。miR-30c-2-3p的高表达与GBM的不良相关,可能是潜在的GBM预后预测分子标记物。

【Abstract】 Objective To investigate the expression profiles of microRNA(miRNA) in glioblastoma(GBM) by microarray technique and explore the potential molecular biomarker for GBM.Methods Plasmas were obtained from 22 cases of GBM and 8 cases of healthy volunteers.Total RNA was extracted by RNA extraction kit and hybridized with miRNA microarrays.The miRNA expression profiles of GBM and healthy volunteers were analyzed and differential miRNA expression was determined.The correlation between differential miRNAs expression and survival was analysed by Kaplan-Meier method.Results MiRNA expression profiling was analyzed.19 miRNAs were identified to be deferentially expressed,in which 3 were overexpressed and 16 were underexpressed in GBM.Kaplan-Meier analysis showed that high expression of miR-30c-2-3p was closely associated with a shorter progression-free survival time(P = 0.016).Conclusion 19 deferentially expressed miRNAs are detected in GBM by microarray method.High expression of miR-30c-2-3p is associated with poor prognosis of patients with GBM.MiR-30c-2-3p may serve as a potential molecular prognosis marker.
