骨髓间充质干细胞对5/6肾切除大鼠肾损伤的保护作用

【摘要】 目的观察骨髓间充质干细胞(BMSCs)对5/6肾切除大鼠肾损伤的保护作用,初步探讨其延缓慢性肾衰竭(CRF)进展的可能作用机制。方法将雄性SD大鼠BMSCs用Hoechst33342标记后注入受体雄性SD大鼠体内。40只受体雄性SD大鼠随机分为四组(每组n=10):假手术组、模型组(5/6肾切除+培养液注射)、贝那普利组(5/6肾切除+贝那普利灌胃)、干细胞组(5/6肾切除+BMSCs移植)。造模后第60天观察肾功能改变;应用酶联免疫吸附(ELISA)方法检测血清血管内皮生长因子(VEGF)浓度,采用免疫组化法检测肾组织VEGF的表达,并观察Hoechst33342标记的BMSCs在受体大鼠肾脏的分布情况。结果 5/6肾切除后60 d可见标记的BMSCs定位于肾组织中。贝那普利组和干细胞组肾功能明显好于模型组(P<0.05);干细胞组血清和肾组织中VEGF表达均高于模型组(P<0.05)。结论 BMSCs可向损伤的肾脏迁移,促进机体分泌VEGF,增加VEGF蛋白在肾脏表达,从而改善肾功能。

【Abstract】 Objective To investigate the effect of bone marrow mesenchymal stem cell( BMSCs) transplatation on renal injury in 5 /6nephrectomy rats,and to explore its possible mechanisms.Methods BMSCs were isolated from male SD rats and labeled with Hoechst33342,and then were injected into male recipient rats.Forty male recipient SD rats were divided randomly into four groups( n =10 in each group) :sham group,model group( 5 /6 nephrectomy + DMEM-F12),benazepril group( 5 /6 nephrectomy + benazepril)and BMSCs group( 5 /6 nephrectomy + BMSCs).At 60 d after modeling,the renal function was observed and serum VEGF concentration was measured by ELISA method.Expression of VEGF protein was measured by immunohistochemical technique.The distribution of Hoechst33342 positive BMSCs in kidney of recipient SD rats was observed by immunohistochemistry.Results At 60 d,the labeled BMSCs were found in kidney tissues in BMSCs group.Renal function was significantly better in benazepril group and BMSCs group than that in model group.The serum VEGF level and the VEGF expression in kidney tissues were higher in BMSCs group than those in model group,respectively.Conclusion BMSCs could transfer to the damaged kidney,promote the secretion of VEGF,and increase VEGF expression in kidney tissue,and finally contribute to improve renal function.
