重组骆驼蓬脂转移蛋白对宫颈癌HeLa细胞化疗的增敏作用
Sensitization of chemotherapy for human cervical carcinoma HeLa cells by recombinant Peganum harmala lipid transfer protein
【摘要】 [目的]研究重组骆驼蓬脂转移蛋白(recombinant Peganum harmala lipid transfer protein,r Ph LTP)与顺铂(DDP)协同对宫颈癌He La细胞的抑制作用及可能的作用机制。[方法]四甲基偶氮咗蓝(MTT)法检测He La细胞生长的抑制率;Annexin-V/PI双染法检测细胞凋亡率,流式细胞术检测细胞内活性氧(ROS)的变化情况。[结果]r Ph LTP与DDP联合应用后与相应的DDP组相比,对细胞生长的抑制率显著增强(P<0.01),低浓度r Ph LTP联合2.5μg/m L顺铂对He La细胞的抑制率为(42.50±0.059)%,远高于高浓度顺铂(10μg/m L)对He La细胞的抑制率(36.88±0.134)%,细胞的凋亡率显著增加(P<0.01),细胞内ROS极明显升高(P<0.001)。[结论]r Ph LTP能够显著增强顺铂对He La细胞的化疗敏感性(P<0.001),这种作用可能与r Ph LTP增强顺铂诱导细胞内活性氧的产生导致细胞凋亡有关。
【Abstract】 [Objective]To investigate the effect of recombinant Peganum harmala lipid transfer protein( r Ph LTP) on the sensitivity of He La cells to cisplatin( DDP) chemotherapy and their possible mechanism of actions. [Methods]The cell growth inhibitory rate was detected through MTT assay in vitro. The cell apoptosis,and the changes of ROS were measured with Annexin- V / PI staining and flow cytometry,respectively. [Results]Compared with individual DDP group,the proliferation inhibition rate( P < 0. 01),The inhibitory rate of low concentration r Ph LTP combine with 2. 5μg / m L DDP was( 42. 50 ± 0. 059) % that was much higher than the inhibitory rate of high concentration DDP( 10μg / m L) which rate was( 36. 88 ± 0. 134) %. And the apoptosis rate( P < 0. 01) were significantly enhanced when He La cells were treated with both r Ph LTP and DDP. Also,the level of ROS generation was increased( P < 0. 001). [Conclusion]r Ph LTP can induce apoptosis in He La cells. Combination of r Ph LTP and DDP displays a synergistic effect( P < 0. 001). The possible mechanism might be that r Ph LTP induce more intracellular ROS generation,which lead to cell apoptosis.
