抗金葡菌深海真菌的分离鉴定及抑菌谱研究
Isolation,identification and antimicrobial spectrum of deep- sea fungi against Staphylococcus aureus
【摘要】 [目的]从深海淤泥样品中分离筛选具有抗金葡菌特性的真菌菌株,对其进行鉴定,并研究其抑菌谱。[方法]采用稀释涂布平板法分离真菌菌株,对峙培养法初筛,琼脂扩散法复筛,将抑菌效果最好的菌株进行形态鉴定、镜检、26S r DNA鉴定,并研究其对产黄青霉、尖孢镰刀菌、黄瓜菌核、木霉、盘单毛孢、平头刺盘孢霉6种真菌和大肠埃希氏菌、单增李斯特菌、副溶血性弧菌、鼠伤寒沙门氏菌4种细菌的抑制作用。[结果]得到1株抑制金葡菌效果最佳的菌株,编号1B12,其抑菌圈直径为35.31 mm,鉴定结果为外瓶霉属(Exophiala),其对上述6种真菌和4种细菌都有一定的抑制作用。[结论]1B12是一株具有广谱抗菌性的海洋真菌。
【Abstract】 [Objective]To isolate a strain of marine fungi against Staphylococcus aureus from deep- sea mud samples and identify it. To study it’s antimicrobial spectrum. [Methods]Strains of fungi were separated by dilution- plate method and selected by primary screening through confronting culture method and second screening through agar well diffusion method. The best inhibiting effect strain was identified by morphological,microscopy and 26 S r DNA identification. The antibacterial spectrum experiments were studied against Pencillium chrysogenum Thom,Fusarium oxysporum Schl,Sclerotinia sclerotiorum,Trichoderma vride Pers,Monochaetia saccardiana( Voglino)Sacc. Traverso,Colletotirchum truncatum,Escherichia coli,Vibrio parahaemolyticus,Salmonella typhimurium and Listeria monocytogenes.[Results]1 strain was screened and selected,named 1B12. The average diameter of the inhibition zone was 35. 31 mm. It belongs to Exophiala. It showed some inhibiting effect against the 6 fungi and 4 bacteria above. [Conclusion]1B12 is a marine fungi which has a broad antibacterial spectrum.
