原生质体融合选育耐高温柠檬酸生产菌

【摘要】 为获得耐高温柠檬酸生产菌,利用原生质体融合技术将具有优良性状的高产菌株W和耐高温柠檬酸生产菌Y-711进行融合,筛选耐高温的高产柠檬酸生产菌。通过变色圈筛选得到融合株R502,在40℃培养72 h就可产酸118.7 g/L,其产酸量比亲本菌株分别高10.5%和21.0%,培养时间分别减少14.3%和7.7%,该菌株遗传稳定。原生质体融合是一种有效获得耐高温柠檬酸生产菌的方法。

【Abstract】 To obtain high temperature resistant critic acid producing strain,the protoplast of high-yield strain W and high temperature resistant mutant strain Y-711 were fused. The obtained fusion strain R502 produced 118. 7 g / L acid after cultivation at 40 ℃ for 72 h. The acid yield of stain R502 increased10. 5% compared with strain W and 21. 0% compared with strain Y-711,respectively. Cultivation time reduced 14. 3% compared to strain W and 7. 7% compared to strain Y-711,respectively. The strain R502 was genetically stable. Protoplast fusion is an effective method to obtain a high temperature resistant citric acid producing strain.
