解螺旋酶RECQL5的功能及其在肿瘤中作用的研究进展
Research Progress on the Function of RECQL5 Helicase and its Role in the Tumor
【摘要】 目前,研究已在小鼠和人类中发现5个Rec Q基因家族成员,分别是Recql1、BLM、WRN、Recql4和RECQL5。其中,RECQL5具有抑制姐妹染色体交换、抑制同源重组修复、阻止双链DNA断裂以及与TOPⅡα的协同作用等功能。最近研究在缺乏RECQL5的Apcmin小鼠小肠和结肠中均发现有肿瘤生长,提示RECQL5蛋白与肿瘤的发生密切相关。此外,RECQL5可能是影响喜树碱(CPT)治疗结直肠癌效果的主要因素,并可能是以伊立替康为基础药物治疗结肠癌效果评估的潜在生物标记物。本文就解螺旋酶RECQL5的功能及其在肿瘤中作用的研究进展进行综述。
【Abstract】 The Rec Q family have five member, which have been found in the mice and human cell, containing Recql1, BLM,WRN, Recql4 and RECQL5. As a member of Rec Q family, RECQL5 has the fuction of inhibiting the sister chromatid exchange(SCE),homologous recombination repair(HRR), DNA double-stranded breaks(DSBs) and Synergy with TOP Ⅱ α, etc. Recent studies have shown that RECQL5 deficiency in Apcmin mice results in a significant increase in the incidence of both the colon and the small intestine tumor, indicated that there must be relationship between RECQL5 and tumor. In addition, RECQL5 may be a major determinant for CPT in the treatment of colorectal cancer and a potential biomarker for irinotecan-based treatment for colon cancer. This article review the research progress on the function of RECQL5 helicase and its role in the tumor.
