金针菇根部多糖的结构特征
Structural characterization of polysaccharides from root of Flammulina velutipes
【摘要】 采用60℃低温热风干燥工艺对金针菇根部进行干燥,粉碎后进行热水浸提、Sevage法除蛋白,并利用Sepharose 2B进行纯化得到金针菇根部多糖FTP-2。采用红外光谱法(IR)、高效液相色谱法(HPLC)、气相色谱法(GC)和扫描电镜(SEM)等方法对FTP-2的结构特征进行测定,结果表明,FTP-2中多糖含量为95.77%,旋光度为+50,相对分子质量为2.60×105 u,单糖组成主要为葡萄糖,以吡喃糖环为主,包含α-和β-2种糖苷键,具有较多的侧链和分枝,在扫描电镜下呈絮状排列。
【Abstract】 Drying the Flammulina velutipes root in hot air oven at 60 ℃. The dried roots were pulverized and sifted through 60 mesh sieve to obtain fine powder. The Flammulina velutipes root polysaccharides were extracted in hot water, removed the proteins with Sevage method, purified by in Sepharose 2B gel column. The structural characterization of FTP-2 was carried out by frared spectroscopy(IR), high performance liquid chromatography(HPLC) instrument, gas chromatography(GC) and scanning electron microscopy(SEM). The results showed that the total carbohydrate content of FTP-2 was 95.77%, optical rotation+50 and molecular weight was 2.60×105 u with many side chain and branch. The monosaccharide composition was glucose mostly and it consisted of pyranose ring with both α-configuration and β-configuration. FTP-2 seemed flocculent at SEM.
