不同干燥方法对菊花褐变相关酶及活性成分的影响研究
Effects of different drying methods on the enzyme activities and active compounds contents from Chrysanthemum morifolium Ramat
【摘要】 目的:研究不同干燥方法对菊花褐变相关酶活性及活性成分的影响,为菊花采后干燥加工提供依据。方法:采用蒸制干燥、恒温鼓风干燥、阴干对菊花的全花和花蕾样品进行干燥处理,利用分光光度计法测定样品多酚氧化酶、过氧化物酶的酶活性以及总酚、总黄酮含量,高效液相色谱法测定绿原酸、木犀草苷和芹菜素含量。结果:蒸制干燥处理的菊花样品,多酚氧化酶及过氧化物酶均可被完全灭活,总酚、总黄酮、绿原酸、木犀草苷和芹菜素含量均高于风干和阴干;干燥处理后的样品总黄酮含量的损失大于50%,总酚类的损失小于50%,不同干燥处理后全花和花蕾的绿原酸含量:蒸制干燥>鼓风干燥>阴干。全花和花蕾中POD活性均与总黄酮含量显著相关,全花中PPO活性与绿原酸含量显著相关。结论:褐变相关酶PPO和POD活性与酚类、黄酮类物质均有一定相关性,蒸制干燥可完全灭活褐变相关酶,其活性成分含量高于风干和阴干。
【Abstract】 Objective: To improve the drying method and the quality of Chrysanthemum morifolium Ramat, the effects of different drying methods on the enzyme activities and the contents of active compounds from the flowers and bud of Chrysanthemum morifolium was studied. Method: Steaming drying method, air blast drying and shade drying were chose to deal with the flowers and bud of Chrysanthemum morifolium. The enzyme activity and the content of total phenoltotal, total flavonoids were determined by spectrophotometer. The contents of chlorogenic acid, luteoloside and apigenin were determined by HPLC. Results: Using the steaming drying method, the activities of polyphenol oxidase and peroxidase were deactivated totally. The contents of total phenolics, total flavonoids, chlorogenic acid, luteoloside and apigenin were highest compared with other two drying methods. The residual contents of total phenolics remained above 50%, and the residual contents of total flavonoids were less than 50%. The contents of chlorogenic acid were: Steaming drying>air blast drying>shade drying. The activities of PPO and POD of flowers and bud of Chrysanthemum morifolium correlated the content of flavonoids substances significantly, the activities of PPO of flowers correlated the content of chlorogenic acid significantly. Conclusion: The enzyme activities of PPO and POD correlated the content of phenolic substances and flavonoids. The activities of polyphenol oxidase and peroxidase were deactivated totally by using the steaming drying method, the contents of active compounds were higher compared with other two drying methods.
