响应面试验优化碱溶酸沉法提取山药储藏蛋白工艺
Optimization of Extraction Conditions for Yam Storage Proteins through Alkali-Extraction and Acid-Precipitation by Response Surface Methodology
【摘要】 以河南焦作白玉山药为原材料,在单因素试验的基础上,采用响应面法中的Box-Behnken试验设计,对影响山药储藏蛋白提取率的3个影响因素即抽提p H值、液料比和抽提时间进行优化。结果表明:各因素对山药储藏蛋白提取率有显著影响,因素影响主次顺序为:抽提p H值>液料比>抽提时间,提取山药储藏蛋白的最佳工艺条件为抽提p H 8.5、液料比3∶1(m L/g)、抽提时间20 min,在此条件下山药储藏蛋白提取率的最大理论值为57.88%,实际蛋白提取率为57.74%。所得山药储藏蛋白提取回归模型显著,拟合性好,可用于预测山药储藏蛋白提取率。
【Abstract】 The extraction of storage proteins from ‘Baiyu’ yam tubers grown in Jiaozuo, Henan province was optimized using combination of one-factor-at-a-time method, Box-Behnken design(BBD) and response surface methodology(RSM). Three factors influencing the extraction yield of yam storage proteins, i.e., p H, liquid/material ratio and extraction time were investigated. The results showed that all these three factors had a notable effect on the extraction yield of yam storage proteins and their significance could be ranked in a decreasing order of p H > liquid/material ratio > extraction time. The optimal extraction conditions were determined as 8.5, 3:1(m L/g) and 20 min for p H, liquid/material ratio and extraction time, respectively. The predicted extraction yield of yam storage proteins under the optimized conditions was 57.88%, agreeing with the experimental value(57.74%). The regression model predicting the extraction of yam storage proteins was highly significant and had excellent goodness of fit, suggesting its usefulness.
