以冬虫夏草与蛹虫草融合株为研究材料,对其原生质体进行紫外诱变,筛选优质高产的新型菌株。得到了高产菌株Y-90,虫草酸产量是出发菌株的1.80倍,高产菌株YP-42,虫草素产量是出发菌株的1.33倍。采用随机扩增多态性DNA(random amplifi ed polymorphic DNA,RAPD)分子标记技术和核糖体r DNA内部转录间隔区(internal transcribed spacer,ITS)序列分析技术对突变株的遗传变异情况进行分析。结果表明突变株的ITS序列并没有发生变化。RAPD结果显示突变株的扩增片段与亲本菌株不同,说明发生了基因突变,其高产性状是可遗传的性状,可以将其用于进一步的育种研究。
