超声空化法对脱细胞血管支架组织结构及生物力学特性影响的实验研究

【摘要】 目的 利用一定强度的超声波,在不影响支架材料本身生物学特性的前提下,使脱细胞血管支架的胶原纤维疏松化,以利于种子细胞的黏附和长入。方法 在无菌条件下取新鲜兔股动脉,反复冻溶+超高压处理后,利用核酸酶进行消化,脱去残留细胞碎片,形成脱细胞血管支架。将不同强度的超声作用于脱细胞血管支架,通过对处理后支架材料的组织结构、生物力学特性的检测和观察,得出最佳的超声处理强度。结果 超声处理参数为强度300 W,处理间隔1 s,处理时长1 min时,脱细胞血管支架材料疏松化程度较未处理前明显提高,而生物力学强度无明显降低。结论 超声空化法可以改善脱细胞血管支架材料结构致密的问题,为组织工程血管支架材料的研究提供新的思路和方法。

【Abstract】 Objective By using a certain intensity ultrasonic to loose the collagen fibers to facilitate seed cell adhesion and ingrowths without affecting the biomechanical characteristics of the scaffold itself.Metho ds Vessels were isolated under sterile conditions from rabbits.After repeated freeze thawing and hyperpressure treatment,the cell debris was taken off to form the acellular vascular scaffold with nucleotidase digestion.Various intensity of ultrasonic was used to loosen the acellular scaffold.Then,tissue structures and biomechanical characteristics of the treated acellular vascular scaffold were observed to find out a optimal ultrasonic intensity.Results With the parameter of 300 watts intensity,1 second interval and 1 minute ultrasonic treatment,we can find a unremarkably changed biomechanics compatibility and more loosen structure in comparing with normal acellular vascular scaffold.Conclusion Ultrasound cavitation method can improve the problems of the dense structure of acellular vascular scaffolds and provides us a new idea and method.
