滋补脾阴方药对脾阴虚痴呆大鼠脑内NMDA受体mRNA表达的影响
Effect of Zi-Bu Pi-Yin Recipe on m RNA Expressions of NMDA Receptor in Different Brain Regions of Spleen-yin Deficiency Alzheimer’s Disease Model Rats
【摘要】 目的:观察滋补脾阴方药对脾阴虚痴呆大鼠脑内N-甲基-D-天冬氨酸受体(NMDAR)亚单位NR1、NR2A、NR2B m RNA表达的影响。方法:运用逆转录聚合酶链反应(RT-PCR)技术,对NMDAR亚单位NR1、NR2A、NR2B m RNA在各脑区的表达状况作相应的检测。结果:痴呆组以及脾阴虚痴呆组大鼠NMDAR亚单位NR1、NR2A、NR2B m RNA在各脑区表达均明显减弱下调(P<0.05);ZBPYR治疗组NMDAR亚单位NR1、NR2A、NR2B m RNA均明显上调(P<0.05)。结论:使用ZBPYR治疗的大鼠NMDAR m RNA的表达水平在各脑区均明显上调,这表明ZBPYR以提升NMDAR m RNA的方式,使得NMDAR蛋白的含量显著增多,而发挥其抗痴呆作用。
【Abstract】 This study was aimed to observe the effect of Zi-Bu Pi-Yin Recipe(ZBPYR) on the m RNA expressions of NMDA receptor subunits NR1, NR2 A, NR2 B in different brain regions of spleen-yin deficiency Alzheimer’s Disease(AD) model rats. The levels of NR1, NR2 A, NR2 B m RNA expressions were detected by using RT-PCR method. The results showed that the levels of NR1, NR2 A, NR2 B m RNA expressions of AD group and spleen-yin deficiency AD group decreased significantly(P < 0.05). The levels of NR1, NR2 A, NR2 B m RNA expressions of ZBPYR treatment group increased significantly(P < 0.05). It was concluded that the expression levels of NMDAR m RNA in different brain regions of the ZBPYR treatment group increased significantly, which indicated that ZBPYR may up-regulate the protein expressions of NMDAR by increasing the expression levels of NMDAR m RNA, thereby to play the anti-dementia effect.
