建立并利用一测多评法优化玄参饮片润透工艺的研究
Optimization of Processing Technology for Scrophularia Ningpoensis by QAMS
【摘要】 目的:建立以一测多评法测定玄参中哈巴苷、肉桂酸、哈巴俄苷含量的方法,利用并验证一测多评法优化玄参饮片润透炮制最佳工艺的可行性和技术适应性。方法:采用一测多评法,以肉桂酸为内标物质,用外标法测定其含量,在一定线性范围内,建立肉桂酸与哈巴苷、哈巴俄苷的相对校正因子,并用该校正因子进行哈巴苷、哈巴俄苷的含量计算,实现一测多评。以加水量、浸润时间、干燥温度为考察因素,采用一测多评法结合外标法测定哈巴苷、哈巴俄苷、肉桂酸含量,比较两种方法测定的结果,并以哈巴苷、哈巴俄苷炮制前后变化率为指标,优化玄参饮片润透工艺。结果:建立的相对校正因子分别为f肉桂酸/哈巴苷=0.060 7(RSD=0.097%)、f肉桂酸/哈巴俄苷=0.304(RSD=0.682%),且差异性较小;玄参最佳润透工艺为加水量为药材0.6倍,浸润时间20 h,干燥温度50℃。结论:建立的一测多评法方法可行,结果可靠;优化了玄参最佳润透工艺,为玄参的润透工艺研究提供了实验依据。
【Abstract】 This study was aimed to establish the quantitative analysis of multi-components by single-marker(QAMS) method for content determination of harpagide, cinnamic acid and harpagoside in Scrophularia ningpoensis. QAMS method was used in the validation of processing technology optimization of S. ningpoensis. QAMS method was used. Cinnamic acid was used as the internal marker. And external marker method was applied in the content determination. Within certain linear range, the related correction factor(RCF) of cinnamic acid to harpagide and harpagoside was established. And RCF was used in the content calculation of harpagide and harpagoside. Water addition, wetting time and drying temperature were used as detection factors. QAMS and external marker method were combined in the content determination of harpagide, harpagoside and cinnamic acid. The detected results by both methods were compared. The change rates of harpagide and harpagoside before and after processing were used as indexes for the optimization of wet technology of S. ningpoensis. The results showed that the established RCF were fcinnamic acid /harpagide=0.060 7(RSD=0.097%) and fcinnamic acid /harpagoside=0.304(RSD=0.682%) with little difference. The best wet technologies of S. ningpoensis were when the water addition of 0.6 times, the wetting time of 20 h, and the drying temperature at 50℃. It was concluded that the established QAMS method was practical with reliable results. This method optimized wet technology and provided experimental evidences for the study on wet technologies of S. ningpoensis.
