火绒草对2型糖尿病大鼠海马神经元Aβ1-42蛋白表达和形态学的影响

【摘要】 目的:观察火绒草水提取物对2型糖尿病(T2DM)大鼠海马区Aβ1-42蛋白表达和形态学的影响。方法:健康雄性Wistar大鼠30只,随机均分为对照组、T2DM组和火绒草组。血糖仪检测大鼠空腹血糖,免疫组化法检测海马CA1区,CA3区,DG区Aβ1-42蛋白的表达,尼氏染色检测海马神经元数量和形态学的变化。结果:T2DM组与对照组比较,空腹血糖明显升高(P<0.01),海马CA1区,CA3区,DG区Aβ1-42蛋白表达较明显增多,海马区神经元数、细胞浆尼氏小体数量减少;火绒草组与T2DM组比较,空腹血糖明显降低(P<0.01),海马区CA1区,CA3区,DG区Aβ1-42蛋白表达明显减少,海马区神经元损伤程度明显减轻,神经元、尼氏小体数量增多。结论:火绒草能降低T2DM大鼠血糖,减少海马区Aβ1-42蛋白表达,对T2DM大鼠神经元损伤有保护作用。

【Abstract】 Objective: To study the effects of Leontopodium alpinum on the Aβ1-42 protein expression and morphology of hippocampal neurons in the hippocampus of type 2 diabetes mellitus( T2DM) rat. Methods: Thirty male Wistar rats were divided into control group,T2 DM group and Leontopodium alpinum group randomly using blood glucose Meter to test blood glucose,immunohistochemical method to detect the expression of Aβ1-42 protein in hippocampus( CA1,CA3,DG)and Nissl staining to examine the number of hippocampal neurons and morphological changes. Results: Compared with control group,T2 DM group showed significantly increased blood glucose value( P < 0. 01) and expression of Aβ1-42 protein in hippocampus was significantly increased and Nissl body was seriously injured with decreased number. Compared with T2 DM group,Leontopodium group in CA1. CA3 and DG regions decreased blood glucose value( P < 0. 01),expression of Aβ1-42 protein in hippocampus was significantly decreased in CA1,CA3 and DG regions,and injury of neurons was reduced obviously showing increased number of Niss bodies and neurons. Conclusion: Leontopodium alpinum can reduce blood glucose value,reduce the expression of Aβ1-42 protein in hippocampus and has a protective effect to neuron of T2 DM rats.
