脾胃气虚型胃溃疡大鼠病证结合模型的制备
Preparation of the Gastric Ulcer Rat Model with Deficiency of Spleen and Stomach
【摘要】 目的:制备胃溃疡脾胃气虚证的大鼠模型,为证型临床客观化、疗效判定规范化奠定基础。方法 :采用"过食酸味加耗气破气"法制备模型,通过行为学观察和检测血清相关指标检验验证造模是否成功。结果:模型组大鼠便溏、脱肛、消瘦较为严重,反证组有所改善;模型组胃蛋白酶活性明显大于空白组,差异具有高度统计学意义(P<0.001),反证组胃蛋白酶活性明显低于模型组,差异具有高度统计学意义(P<0.01);模型组MDA含量高于空白组及反证组,但与空白组比较无显著性差异,与反证组有显著性差异(P<0.05);模型组胸腺湿重和指数、NO、6-keto-PGF1α含量及SOD活性明显低于空白组,分别具有不同显著性差异(P<0.01,P<0.05,P<0.001,P<0.001),反证组该四种指标明显高于模型组,分别具有不同显著性差异(P<0.05,P<0.05,P<0.001,P<0.01)。结论 :采用"过食酸味加耗气破气"法制备的脾胃气虚证胃溃疡模型符合临床症状表现,验证胃溃疡"脾胃气虚"型大鼠模型成功。
【Abstract】 Objective:To prepare the gastric ulcer rat model with deficiency of spleen and stomach,so as to lay the foundations for clinical syndromes objectification and standardization effect judgment. Methods:Using the method of eating sour excessive food plus destructing and consuming righteousness to prepare rat model,by observing the behavior and testing related indices in serum of rat model to verify whether it was made successfully. Results:Rats in the model group appeared prolepses of the anus,loose stools and weight loss seriously. The situation of disproof group improved. The protease activity of rats in the model group was sig nificantly higher than that of the blank group(P<0.001),and the protease activity of disproof group was significantly lower than that of the model group(P<0.01). The Malondialdehyde(MDA) content of model group was higher than those of the blank group and disproof group,no significant differences compare with blank group,but there was significant differences compare with disproof group(P<0.05). The weight and index of thymus,the content of nitric oxide(NO),6-keto-PGF1α and superoxide dismutase(SOD) activity in model group were all significantly lower than those of blank group(P<0.01,P<0.05,P<0.001,P<0.001),and these indices of disproof group were all significantly higher than those of model group(P<0.05,P<0.05,P<0.001,P<0.01). Conclusion:Using the method of eating sour excessive food plus destructing and consuming righteousness to prepare the gastric ulcer rat model with deficiency of spleen and stomach was consistent with clinical symptoms. It indicated that the models have been completed successfully.
