高压氧对大鼠脊髓损伤的治疗作用及机制

【摘要】 目的观察高压氧(HBO)治疗对大鼠脊髓损伤(SCI)的治疗作用,并探讨其可能机制。方法 25只健康清洁级成年SD大鼠随机分为SCI组10只、HBO组10只、对照组5只,SCI组、HBO组大鼠均制备SCI模型,HBO组制模成功后采用HBO治疗;对照组采取同前方法咬开椎板,不损伤脊髓,后给予缝合。观察两组大鼠不同时点(制模成功后6 h及1、3、7、14、21 d,分别计为T1、T2、T3、T4、T5、T6)BBB评分及脊髓组织中突触素ⅠmRNA。结果与对照组比较,HBO组、SCI组各时点(T3、T4、T5、T6)BBB评分及脊髓组织突触素ⅠmRNA表达升高(P均<0.05);与SCI组比较,HBO组各时点(T3、T4、T5、T6)BBB评分及脊髓组织突触素ⅠmRNA表达升高(P均<0.05)。结论 HBO可促进大鼠损伤脊髓的恢复,其机制可能与上调损伤脊髓组织内突触素ⅠmRNA表达有关。

【Abstract】 Objective To observe the therapeutic effect of hyperbaric oxygen( HBO) on the spinal cord injury( SCI),and to explore its possible mechanism. Methods Twenty-five healthy and clean SD adult rats were randomly divided into the SCI group( n = 10),HBO group( n = 10) and the control group( n = 5). The SCI model rats were prepared in the SCI and HBO groups,after the success of molding,the HBO group received HBO treatment; the vertebral plates of rats in the control group were opened and sutured without damage to the spinal cord. BBB rating and synaptophysin Ⅰ mRNA in the spinal cord tissues at different time points( 6 h after molding and 1,3,7,14 and 21 d,which is counted as T1,T2,T3,T4,T5 and T6) were observed in the two groups. Results Compared with the control group,the BBB rating and synaptophysin Ⅰ mRNA expression was increased in the HBO group and SCI group at each time point( T3,T4,T5 and T6)( all P < 0. 05). Compared with the SCI group,the BBB rating and synaptophysin Ⅰ mRNA expression was increased in the HBO group at each time point( T3,T4,T5 and T6)( all P < 0. 05). Conclusion HBO promoted the recovery of the injured spinal cord of rats,which may be related with the up-regulation of synaptophysin Ⅰ mRNA expression in the injured spinal cord tissues.
