复方灵芝胶囊中粗多糖测定方法研究

【摘要】 目的建立复方灵芝胶囊中粗多糖的含量测定方法。方法将复方灵芝胶囊内容物用糖化酶酶解,醇沉法提取粗多糖,苯酚-硫酸法测定其含量,并对糖化酶用量、酶解时间条件进行了优化。结果糖化酶用量0.1 g,酶解时间60 min为优,测定粗多糖的标准曲线相关系数为0.9995,平均回收率为98.41%,RSD为3.50%。结论该方法操作简便,显色稳定、重现性及稳定性良好,适用于复方灵芝胶囊中粗多糖的含量检测。

【Abstract】 Objective To establish a method for determination of the content of crude polysaccharide in Compound Ganoderma Capsules. Methods The contents of Compound Ganoderma Capsules were hydrolyzed by glucoamylase, the crude polysaccharide was extracted by alcohol precipitation, and the content of the crude polysacharide was determined by phenol-sulfuric acid method. The dosage of glucoamylase and the enzymolysis time were optimized. Results The optimized enzymolysis conditions were as follows: glucoamylase dosage of 0.1 g and hydrolysis time of 60 min. The correlation coefficient of the standard curve for determination of crude polysaccharide by this method was 0.9995, the average recovery rate was 98.41 %, with RSD of 3.50 %. Conclusion This method is easy to operate, also shows good repeatability and stability, and is suitable for detection of the content of polysaccharides in Compound Ganoderma Capsules.
