口腔炎合剂急性毒性和免疫功能影响试验研究

【摘要】 目的观察口腔炎合剂的急性毒性,同时研究口腔炎合剂对小鼠非特异性免疫功能的影响。方法受试药以最大浓度,最大灌胃体积经口给予小鼠,测其经口给药的最大耐受量;采用腹腔注射环磷酰胺诱导免疫低下小鼠模型,口腔炎合剂按低、高剂量组灌胃,连续14d。称体重、胸腺重和肝脾重,计算胸腺指数、脾指数、炭粒廓清指数K和吞噬指数α等,并进行统计学处理。结果最大浓度、最大灌胃体积下灌胃给予小鼠,动物无死亡,受试药小鼠最大给药量相当于成人常用量的168倍。口腔炎合剂高剂量组能提高该模型小鼠的脾指数,差异有统计学意义(P<0.05)、廓清指数K和校正吞噬指数α(P<0.05);口腔炎合剂低剂量组对该模型小鼠炭粒廓清指数K、校正吞噬指数α无明显影响(P>0.05)。结论口腔炎合剂在常规剂量下使用是安全的。口腔炎合剂高剂量对环磷酰胺诱导免疫低下小鼠有提高非特异性免疫的作用,具有一定的免疫调节作用。

【Abstract】 Objective To observe the acute toxicity of stomatitis mixture at the same time study stomatitis mixture effect on nonspecific immune function in mice. Methods The reagent with maximum concentration,the maximum to fill the stomach through the mouth volume for mice,measuring its through the mouth for maximum tolerance; Induced by intraperitoneal injection of cyclophosphamide low immune model mice,stomatitis mixture at low and high dose group to fill the stomach,for 14 consecutive days,at the same time,according to body weight,thymus weight and liver and spleen weight,thymus index,spleen index,K carbon clearance index and the index of alpha,etc.,and carry on statistics processing. Results Gastric lavage under the volume of the maximum concentration,giving mice,animal without death,dosage of mice by reagent dosage is equal to the largest adult often168 times. Stomatitis mixture high dose group can improve the model of mice spleen index( P < 0. 05),expurgation index K index and calibration of alpha( P < 0. 05); Stomatitis mixture low dose group of the carbon clearance index K,correction model mice consuming index had no significant effect of alpha( P > 0. 05). Conclusion Stomatitis mixture used in conventional doses are safe. Stomatitis mixture of high dose of cyclophosphamide induced low immune mice have improve the non-specific immune function,immune regulation.
